Objective: To explore the relation between advanced glycosylation end products and ratosteoblasts osteogenic capabilities by observing Effect of advanced glycosylation endproducts (AGES) On Expression of BMP-2and ALP in osteoblasts rats.Methods: Isolated and cultured rat osteoblasts spread to four generations,identified calciumnodule formation of osteoblasts by alizarin red staining.Control group and5mg/mladvanced glycosylation end products(AGES) at a dose of50ul,100ul,150ul experimentalgroups.Advanced glycosylation end products(AGES) after treatment for24hours, theexpression of BMP-2mRNA is detected by Real-Time PCR, BMP-2and ALP weredetected by Elisa.Result: Advanced glycosylation end products (AGES)can decrease the expression levels ofBMP-2mRNA,and decrease the expression of BMP-2protein levels and ALP.Conclusion: AGES could significantly inhibit the formation of bone, The mechanism maybe related to effects of AGES on down-regulated expression of BMP-2and ALP. |
