Objective: To investigate the expression of leukocyte-associated Ig-likereceptor-1(LAIR-1) in childhood immune thrombocytopenia(ITP) and explore the possiblerole on pathogenesis in childhood ITP.Methods: Using flow cytometry detected the expression of CD4+T cells, CD8+T cellsand CD19+CD20+B cells and the expression of LAIR-1on CD4+T cells, CD8+T cells andCD19+CD20+B cells from peripheral blood of children with ITP; Using ELISA (doubleantibody sandwich method) detected serum LAIR-1level; Using Real time PCR detectedthe expression of LAIR-1mRNA.Results:40children with ITP and32health children (as control group) were enrolled inthis study.(1)The ratio of CD19+CD20+B cells were significantly higher (P<0.05) compareto control group, while the ratio of CD4+T cells were significantly decreased in ITP group(P<0.05). The ratio of CD8+T cells increased in ITP group but with no significance(P>0.05).(2)The expression of LAIR-1on CD4+T cells, CD8+T cells were significantlylower in ITP group compare to control group (P<0.05), and the expression of LAIR-1onCD19+CD20+B cells was decreased, but with no significance compare to control group(P>0.05).(3)The sLAIR-1level significantly increased in ITP group to control group(P<0.05).(4)The level of the LAIR-1mRNA in ITP group was significantly increased thancontrol group (P<0.05).Conclusion: The expression of CD4+T cells were decreased and CD19+CD20+B cellswere increased, which can suggest the presence of immune imbalance in ITP.LAIR-1onCD4+T cells,CD8+T cells were decreased, and the level of sLAIR-1in serum wereincreased, which suggest LAIR-1and sLAIR-1may play an important role in immuneimbalance in childhood ITP.
