Study On Chemical Constituents Of Schisandra Sphenanthera And Marine-derived Fungi Aspergillus Carneus

To study the chemical constituents of Schisandra sphenanthera Rehd. Et Wils. andsecondary metabolites from marine-derived fungi Aspergillus carneus HQ889708.Twenty-five compounds (including there novel ones) were isolated and purified by usingvarious chromatographic methods (silica gel column chromatography, sephadex LH-20chromatography and preparative HPLC). Structures were elucidated by extensivespectroscopic analysis (including1D and2D-NMR, MS, OR, ECD and VCD).Sixteen compounds were obtained from the ethanol extracts of fruits of Schisandrasphenanthera Rehd. Et Wils. The strctures were identified as four simple phenylpropanoids:methyl caffeate (hp-01), Methyl phydroxycinnamate (hp-02), trans-cinnamic acid (hp-03),methyl caffeate-β-D-glcoside (hp-04); Two simple lignans:(-)-isolariciresinol (hp-05),(+)-secoisolariciresinol (hp-06); Three dibenzocyclooctene lignans: Schizandrin (hp-07),angeloyl-schizandrin (hp-08), gomisin A (hp-09); Two dihydrochalones: trilobatin (hp-10),phloridizin (hp-11); Two flavanones: naringenin-7-O-β-D-glucoside (hp-12) and huazhongilexone-7-O-β-D-glucoside (hp-13); Two steroids: β-sitosterol (hp-14), daucosterol (hp-15);Other types: methyl gallate (hp-16). Among above compounds, hp-08and hp-10~13wereisolated from this genus for the first time. In addition, by careful vibrational circulardichroism (VCD) investigation of (7S,8S) and (7S,8R) using B3LYP/6-311+G(d) methods,absolute configuration of (+)-schizandrin is assigned as (+)-(7S,8R).Nine compounds were obtained from the fermentation broth of A. carneus HQ889708,which was isolated from seawater (South China Sea), including three novel phenyl etherderivatives:3-hydroxy-5-(3-hydroxy-5-methylphenoxy)benzoic acid (AC-05),3,4-dihydroxy-5-(3-hydroxy-5-methylphenoxy)benzoic acid (AC-07),3-(3-hydroxy-5-(hydroxymethyl)phenoxy)-5-methylphenol (AC-09); Three dipeptide:1,4-benzodiazepine-2,5-diones (AC-03),cyclopenin (AC-04), dehydrocyclopeptine (AC-06); sterigmatocystin (AC-01), viridicatin(AC-02), viridicatol (AC-08), all above are obtained from the strain for the first time.
