Constipation is one of common clinical digestive diseases, it affects the patients’ appetite and intestinal absorption of nutrients, it is one of the important factors of inducing or aggravating of colorectal cancer, cardiovascular and cerebrovascular disease. With the dietary change and increasing social pressure, constipation and the incidence are increasing every year, and it seriously affects the quality of people’s life.Traditional Chinese treatment of constipation usually base on the practioner’s own clinical experience to differentiation and Herbal Prescription, and resulting in diverse and even random methods of treatment. There are many compounds which have the laxative effect. Based on the experience of clinicians, this research did addition and substraction on Runchang Pills, and developed a new type of laxative prescription-Mitong capsule by Jiudahuang, hemp seed, angelica and glutinous rice (fried). Jiudahuang is purgative and activating, it can eliminating heat and purgating fire, complemented by the hemp seed and angelica that moistening and skidding the intestine, enriching blood and helping blood circulation, combined with fried glutinous rice with the benefit of replenishing qi, spleen and stomach the whole combination of the drugs can purgate stagnation, enrich blood and moisten intestines.It is for the treatment of excess heat, constipation, intestinal dry constipation, suitable both for senile habitual constipation and young hot junction constipation disease.With the orthogonal design method, the Jiudahuang ethanol extraction process and hemp seed and Angelica ethanol extraction process were optimized. Use1,8dihydroxyanthraquinone as the reference, the spectrophotometric detection of Jiudahuang content to select three factors:the ethanol concentration, ethanol consumption and extraction time; and three levels of each factor, orthogonal design test choose the best extraction process five times the amount of80%by ethanol extract three times (2,1,1hr). Using the fat content of hemp seed and Angelica for the evaluation to select the ethanol concentration, ethanol consumption and extraction time factor, three levels of each factor, the orthogonal design method to the optimal extraction process is the way that4times the amount of90%ethanol reflux extiraction three times (3,2,1hr).The preparation process of the Mitong capsules is to crush and sieve (20mesh,40mesh,40mesh) Jiudahuang, hemp seed and Angelica. Jiudahuang to five times the amount of80%ethanol reflux extraction three times (2,1,1hr), filtration, the combined filtrate, ethanol recovery, dubbed1g/mL Jiudahuang solution, add1.5times the amount of4%gelatin solution to remove the tannins in the wine rhubarb composition, filtration, the filtrate by adding four times the amount of95%ethanol in addition to the excess gelatinrecovery of ethanol, concentrated to a thick paste (A); hemp seed and Angelica to four times the amount of90%ethanol reflux extraction three times (3,2,1hr), filtered, the combined filtrate, ethanol recovery, concentrated to a thick paste (B); merge the two thick paste, then add the rice powder (fried,80mesh), mixe dry powder, granulate, sieving, encapsulated to get Mitong capsule as the final product.
