Background:At present, Lung cancer is a kind of disease that seriously threathuman health. It could be prone to transfer to the brain, the adrenal and otherplaces. And it will influence the quality of life.Lung cancer transfer is a dynamic,multi-step process, mainly divided into three stages: start, progress, engraftmentand produce toxic effect. In the process of lung cancer lymph node metastases,Vessel endothelium growth factor-C,Vessel endothelium growth factor-D effectVEGFR-3(Flt-4) receptor on the lymphatic endothelial surrounding the cancercells by the means of auto-crine chemokine. Synergistic effect of both VEGF-C、VEGF—D and VEGFR-3promotes lymph node metastases of lung cancer. In theprocess of lymphatic transferring of tumors, many proteins are involved in, suchas RhoC, osteopontin (OPN), membrane protein AI; and many genes, such asRhoC, LOX, COX-2, MMP-1RANKL, CTGF. The proteins and the expressionof genes provide the basis for clinical judgment on cancer metastases. AnnexinAI (ANXA1, lipocortinI) is found earliest among the Annexin family members.According to the studies, it is involved in inflammation, cell proliferation anddifferentiation, control signal of cell death, cell apoptosis, and many other life activities. In recent years, research shows that the expression of ANXA1disorder is closely drelated to tumor invasion and metastasis, and the expressionwith tissue specific. AnnexinA1high expresses in the stomach, colonadenocarcinoma, esophageal, pancreas, ductal adenocarcinoma of prostatecancer. And it doesn’t expresse in the liver cells, gastrointestinal glandularepithelium, and tubular cells. AnnexinA1down regulate in breast cancer,cervical squamous cell carcinoma.Objective:To explore the role of Annexin A1in lung cancer with bone metastasis.Methods:1. Totally64cases of clinical lung cancer paraffin specimen from June2009toJune2010in bing li-ke were enrolled in this study(including20cases squamous cellcarcinoma,21cases gland cancer and23cases small cell lung cancer), And groupingaccoring to the clinical diagnosis.2. The immunohistochemical SP ldpe-g-nvp detection lung cancer and lungcancer bone tissue AnnexinA1protein expression. And through the Mann-Whitney Uinspection, compares the between groups Kruskal-Wallis H inspection, AnnexinA1expression of positive between groups Fisher compares the precise the probability methodof statistical analysis. Discusses the expression and difference of research.3. By immune cells chemical method to detect AnnexinA1expression betweenlung cancer cell lines with bone metastasis SBC-5and non-bone metastasis SBC-34. To detect AnnexinA1expression in mRNA and protein in SBC-5cell line andSBC-3cell line by RT-PCR and Western-blot.Results: AnnexinA1has high expression in lung cancer tissue with bonemetastasis and SBC-5cell line, than which in lung cancer tissue without bonemetastasis and SBC-3cell line.The rate of expression of AnnexinA1in lungcancer tissue with bone metastasis is97.1%(33/34);the rate in lung cancer tissue without bone metastasis is43.3%(13/30),the difference havestatistic value (P<0.05).Conclusions: The expression of AnnexinA1may correlate with lung cancerbone metastasis.
