Abstract/Summary:
Objective: To assess the in vitro effects of Azithromycin、Erythromycin、Sulfadiazine and Allitride on Toxoplasma gondii.Methods: The cultures of mouse macrophages and HFF were prepared on glass coverslips in dishes and grown to confluence. Then confluent monolayers were infectedwith~105parasites. Antimicrobial agents at various concentrations were added to thedishs at the same time when tachyzoites were added into the dishs or after tachyzoitesinvade into the macrophages. Cultures were examined at frequent intervals with aninverted microscope to document the infection of cells and the formation ofpseudocysts. The glass cover slips were taken out from the dishes at1、4、8、12、16、20、24、48、72h respectively, washed immediately in Hank’s, stained withGiemsa,and examined microscopically. Results: The inhibitory concentration were20μg/mL for Azithromycin. At40μg/mL80μg/mL Azithromycin, cytotoxic effecton the host cell and parasiticidal effect could be detected by microscopy. At100μg/mL、150μg/mL. Erythromycin were found to have important inhibitory effects on T. gondii.But it is not parasiticidal. At100μg/mL、200μg/mL Sulfadiazine cannot inhibits thegrowth of the T. gondii. And at20μg/mL、40μg/mL Allitride cannot inhibits thegrowth of the T. gondii. Conclusion: The results of this experiment revealedthat Azithromycin have a strong parasiticidal effect on T. gondii. At100μg/mLand150μg/mL Erythromycin cannot kill the parasities, but can inhibite the growth ofT. gondii. At100μg/mL、200μg/mL Sulfadiazine cannot inhibite the growth of T.gondii. At20μg/mL、40μg/mL Allitride cannot inhibite the growth of T.gondi too.
