Objective:To optimize the composition of Haikangling, investigating the protective effect of Haikangling on the damage of cultured SH-SY5Y cells by β-amyloid peptide and its mechanisms; proving the improvement of Haikangling,the compound traditional Chinese medicine on β-amyloid peptide-induced amnesia in rats. Provide a valid experimental data and theoretical basis for the clinical application of Haikangling’s treatment about Alzheimer’s disease,and for traditional Chinese medicine’s multi-target mechanisms.Methods:Part1:Optimized the composition of Haikangling:The damage of cultured SH-SY5Y cells was made by hydrogen peroxide. The protective effects of single herb in Haikangling and Haikangling compound were observed by colorlmetric MTT assay.Part2:The protective effect of Haikangling on the damage of cultured SH-SY5Y cells by β-amyloid peptide and its mechanisms:The damage of cultured SH-SY5Y cells was made by β-amyloid peptide. The protective effects of the new compound of Haikangling were observed by colorlmetric MTT assay.Part3:The new compound of Haikangling affects amnesia in rats induced by β-amyloid peptide.The amnesia in rats was made by β-amyloid peptide.The influence of Haikangling on the amnesia in rats induced by β-amyloid peptide were observed by Morris water maze. Detecte the protective effects of the new compound of Haikangling on the amnesia in rats induced by β-amyloid peptide.Results:Part1:The new compound of Haikangling (0.159g·ml-1)can relieve the damage of SH-SY5Y cells induced by hydrogen peroxide and increase the survival of the cells (P<0.01).Part2:The new compound of Haikangling (0.159g·ml-1)can relieve the damage of SH-SY5Y cells induced by β-amyloid peptide and increase the survival of the cells (P<0.01). The new compound of Haikangling can effectively inhibit neuronal apoptosis induced by β-amyloid peptide.Part3:The results of morris water maze test showed that the rats taking the new compound of Haikangling were significantly shorter escape latency and swimming distance in90s.The experiment of Space exploration showed that the rats swimming time was prolonged on the former platform quadrant. The time that first to the former platform was significantly shorten. Obviously increase the number of crossing the former platform.The Biochemical indices of brain tissue in rats showed that the rats pretreated with Haikangling30days were significantly reduced MDA,and increased the activities of SOD and GSH-Px and also enhanced the AchE activity. The Blood biochemical parameters showed that the rats pre-given Haikangling were significantly decreased the TG and T-CHO and increased the HDL-C in blood. These results suggest that Haikangling has the roles of lowerring blood lipids and preventing atherosclerosis.Conclusion:The new compound of Haikangling can strongly protect the cultured SH-SY5Y cells from damage induced by hydrogen peroxide and β-amyloid peptide.The new compound of Haikangling can improve the learning and memory ability of the amnesia in rats induced by β-amyloid peptide. Its mechanism is relevent with resistance to oxidative damage.
