| Background and objectiveCoronary atherosclerotic artery disease is a multiple genes disease caused by a variety of interaction factors.It has become the major disease which threatens human health in the world today. The pathology base is a vascular luminal stenosis or blockages in myocardial cell damage caused by coronary artery disease. So to study the risk factors that can influence the coronary artery disease have important clinical significance. The traditional risk factors have obtain a relatively detailed study of the evidence,like age, obesity, hyperlipidemia, smoking, hypertension and so on. To study the relationship between gene polymorphism and coronary artery disease also become the focus of research at home and abroad. This study mainly analysis the endothelial type of nitric oxide synthase (eNOS) gene G894T polymorphism by gene chip technology to research the correlation between eNOS gene polymorphism and the severely of coronary artery disease on the old people in Qingdao.MethodsA case-control study was performed to study the association between eNOS polymorphism and coronary artery disease by gene chip technology.100patients having coronary artery disease were scheduled to undergo muliti-slice spiral computed tomography coronary angiography(CTA) and count coronary artery calcium score(CACS). According to CACS to divided into three groups:group A(CACS=0-10), group B(CACS=11-400), group C(CACS>400).Inquiry and check the tradional risk factors that influence the coronary artery disease, such as the history of hypertension, diabetes, hyperlipidemia, family history of coronary heart disease, smoking history and so on. Monitor blood pressure, blood glucose, blood fat periodically for patients.Results1.To identify the gene type of eNOS in the selected patients, the cases of GG, GT, TT genetypes were75,21,4. It was no significant difference according to the Hardy-Weinberg equilibrium test. The group was in H-W equilibrium state(χ2=2.418, P>0.05).2.The GT+TT genetype was different in the patients for the coronary artery calcium score, the high-risk group(group C) was higher than the other two (χ2=21.907, P=0.000).3.Logistic regression was used to assess interactions between eNOS G894T genotypes,smoke and coronaty artery disaease.4. Linear Regression displayed GT+TT genotypes,smoke,hyperlipaemia are influncing factors of coronary artery calcium score(Beta=0.476,0.217,0.191,P<0.05).5.Analysis of covariance excluded the influence of hyperlipidemia and smoking history, the CT genetype still for coronary artery disease risk factors (P<0.01).Conclusions1.There is an association among eNOS G894T genotype, smoking history, hyperlipaemia and coronary artery calcium score..2.eNOS G894T genotypes may be the risk factor for coronary artery disease. |
