Objective To observe the expression of CXCL1in early Aspergillus fumigatus keratitis in a rats model.Materials and methods66Wistar rats were randomly divided into3groups. Group1was normal control group (12eyes), group2was surgical control group (30eyes), group3was fungal infected group(30eyes). After scraping the epithelium of central cornea, Aspergillus fumigatus-laden contact lens were covered on the right eyes of rats in group3, while eyes in groups2were covered with contact lens without fungi. The expression of CXCL1mRNA was detected by RT-PCR4,8,16,24h post infection. The level of CXCL1protein in cornea was tested24h post infection by immunohistochemistry.Results Histopathologic examination showed tissue necrosis and neutrophil infiltration in Aspergillus fumigatus infected cornea. CXCL1mRAN expression increased4h post infection and remain to increase during a24h period post infection (P=0.000). There was CXCL1protein in normal rat cornea, which increased after cornea injury (P=0.000). At24h post infection, CXCL1protein level of the infected group was significantly higher than that of the control group (P=0.000).Conclusion1.The rats’model of Aspergillus fumigatus keratitis is successfully established.2. The main leukocytes infiltrating in Aspergillus fumigatus keratitis is neutrophil.3. CXCL1expression is increased in early Aspergillus fumigatus keratitis in rats.
