The Expression Of Runx3ã€TGF-β1ã€Î²-catenin And Cyclin D1in Primary Liver Cancer And Their Correlation Studies | | Posted on:2013-03-01 | Degree:Master | Type:Thesis | | Country:China | Candidate:W Zhao | Full Text:PDF | | GTID:2284330362972454 | Subject:Surgery | | Abstract/Summary: | PDF Full Text Request | | Objecrive The subject is to detect the expression of Runx3ã€TGF-β1ã€Î²-catenin and CyclinD1in Liver tissues and their corresponding adjacenttissues and normal liver tissues,and to analyze their correlations betweenthem.To have a preliminary study of them in PHC occurrenceã€developmentã€invasionã€metastasis etc,and to understand the interactions between theirrole of signaling pathways.Mehods Two-step non-biotin immunohistochemical method was performed toseparately detect the expression of Runx3ã€TGF-β1ã€Î²-catenin and CyclinD1protein in45PHC tissues and their corresponding adjacent tissues and45normal liver tissues.Results (1) The positive expression rate of Runx3in Liver tissues was35.6%(16/45),the expression rate was significantly lower than thecorresponding adjacent tissues60%(27/45) and normal livertissues80%(36/45),the difference between then was significant(P all<0.05).The abnormal expression of Runx3was associated with portal vein tumorthrombus,clinical stage, capsule and lymph node metatasis(P<0.05),and hadno correlations with genderã€ageã€AFPã€HBsAgã€histological grade and tumorsize(P>0.05).(2) The positive expression rate of TGF-β1in Liver tissueswas40%(18/45),the expression rate was higher than the correspondingadjacent tissues20%(9/45) and normal liver tissues4.4%(2/45),the difference between then was significant(P all<0.05).The abnormal expression of TGF-β1was associated with histological typesã€portal vein tumor thrombusã€lymph node metatasisã€capsuleã€AFP and tumor size(P<0.05),and had nocorrelations with genderã€ageã€clinical stage and HBsAg (P>0.05).(3) Thepositive expression rate of β-catenin in Liver tissues was51.1%(23/45),theexpression rate was higher than the corresponding adjacenttissues26.7%(12/45) and normal liver tissues8.9%(4/45),the differencebetween then was significant(P all<0.05).The abnormal expression of β-catenin was associated with portal vein tumor thrombusã€capsuleã€tumor stageã€intrahepatic or distant lymph node metatasis and HBsAg(P<0.05),and had norelationship with genderã€ageã€tumor diameter〠histological types and AFP(P>0.05).(4) The positive expression rate of CyclinD1in Liver tissues was87.5%(21/45),the expression rate was significantly higer than thecorresponding adjacent tissues24.4%(11/45) and normal livertissues6.7%(3/45),the difference between then was significant(P all<0.05).The abnormal expression of CyclinD1was associated with portal veintumor thrombusã€clinical stageã€histological typeã€lymph node metatasis andHBsAg(P<0.05),and had also no relationships with genderã€ageã€tumor sizeã€capsule and AFP(P>0.05).(5) The expression of Runx3ã€TGF-β1ã€Î²-cateninand CyclinD1were all associated with portal vein tumor thrombus and lymphnode metatasis(P<0.05).(6) The protein expression between any two ofRunx3ã€TGF-β1ã€Î²-catenin and CyclinD1were related:Runx3and TGF-β1:X~2=23.341, P=0.000ï¼›Runx3and β-catenin:X~2=10.405, P=0.001ï¼›Runx3andCyclinD1:X~2=7.774, P=0.005ï¼›TGF-β1and β-catenin:X~2=10.020, P=0.002ï¼›TGF-β1and CyclinD1:X~2=16.205, P=0.000;β-catenin and CyclinD1:X~2=14.032, P=0.000,(Pall<0.05).Conclusions The expression correlation of Runx3ã€TGF-β1ã€Î²-cateninand Cyclin D1implied that:their role of different signaling pathways existinteractions,may co-involved in the occurrence of PHC〠developmentã€progression and metastasis.To have a combined detection of their expressionin PHC can contribute to the early diagnosis and prognosis of PHC,and mayprovide gene therapeutic targets for the treatment of PHC molecular biology. | | Keywords/Search Tags: | Primary liver cancer, immunohistochemistry, Runx3, TGF-β1, β-catenin, Cyclin D1 | PDF Full Text Request | Related items |
