ObjectiveTo investigate the effect of short term hyperglycemia on the expressionof intercellular adhesion molecule-1(ICAM-1) in umbilical vasculartissue of gestational diabetic women.Methods1、GroupingWe enrolled21patients with GDM who received antenatal andobstetric care at the Second Affiliated Hospital of Guangzhou MedicalCollege between April2011and February2012. Twenty-two normalpregnant women were taken as the control group. The age, gravidity,pregnant week, and the history of delivery were not significantly differentbetween two groups,and they did not have any history of smoking or anyother systemic diseases.Between the gestational weeks24and28, all patients were screenedfor GDM by a50g glucose load, and the diagnosis of GDM was madefollowing a75g glucose load. 2、 Specimen preparationSamples were collected in the early postpartum period. They were5cm from maternal side of umbilical cord. After cleaning by cold salinerinse and drying by filter paper, they were placed in10%neutralformaldehyde. Then paraffin-embedded and4μm specimens were takenfor Immunohistochemical staining.3、 Detecting indicatorsICAM-1were tested by Two-step methed immunohistochemicalstaining. Randomized BG and HbA1c were measured routinely.4、 Result judgmentICAM-1expressed in the endothelial cell membrane. Each case wasrandom selected5vision by two viewers. The staining intensity and thepositive rate were recorded. Zero,1,2and3represented colorless, yellow,brown and tan, respectively. Positive rate of staining cells of≤5%,6%-20%,21%~50%,and≥51%were recorded as0,1,2and3points,respectively.ICAM-1expression equaled staining intensity*Positiverate of staining cells. ICAM-1expression0,1~2,3~5and6~9meantnegative(-), weakly positive(+), moderate positive(++) and strongpositive(+++), respectively.5、 Statistical AnalysisSPSS16.0application package was used for the analysis. Student’s Ttest, Mann-Whitney U test and Pearsonχ2test were used. P<0.05wasconsided as significant. Results1、ICAM-1was expressed in the cell membrane of endothelial cells.The ICAM-1expression was not different between GDM group andcontrol group in both umbilical artery and vein(P=0.52; P=0.36).ConclusionIn conclusion, our findings do not support the hypothesis that increasedexpression of ICAM-1in umbilical vascular tissue occurs in gestationaldiabetic women when compared with normal controls.
