This paper consisted of five chapters. Three major investigated topics were covered: firstly, the new synthesis of 1,2,3,5-tetra-O-acetyl-β-D-ribofuranose was studied; secondly, the new method of methyl 1-(2,3,5-tri-O-acetyl-β-D-ribofuranosyl)-1H-1,2,4-triazole-3-carboxylate was explored; lastly,“one pot”preparation of ribavirin with 1,2,3,5-tetra-O-acetyl-β-D -ribofuranose as start material was developed.In chapter one, the progress and classification of antiviral drugs was reviewed, then the research and development strategy of nucleoside antiviral drug were introduced. In chapter two, the synthetic route of ribavirin was reviewed. There was a typical experimental process of operation in chapter three, 1,2,3,5-tetra-O-acetyl-β-D-ribofuranose was prepared by the catalytic acetylation of nucleosides; then the desired acylprotected ribavirin derivative, methyl 1-(2,3,5-tri-O-acetyl-β-D -ribofuranosyl)-1H-1,2,4-triazole-3-carboxylate was synthesized by the condensation with 1,2,3,5-tetra-O-acetyl-β-D-ribofuranose and methyl 1,2,4-triazole-3-carboxylate; finally, ribavirin was obtained by deprotection of ribavirin derivative with subsequent ammonolysis in“one pot”. In chapter four, the experimental results and phenomena of 1,2,3,5-tetra-O-acetyl-β-D-ribofuranose and methyl 1-(2,3,5-tri-O-acetyl-β -D-ribofuranosyl)-1H-1,2,4-triazole-3-carboxylate were discussed from various effects included kinds of the catalyst, amount of the catalyst, reaction feed molar ratio, reaction temperature and time, recrystallization solvents. Conclusions and Outlook of research were depicted in chapter five.Comparing with the old methods, the new ones have the advantages of mild reaction conditions, simple operations, short technology process, high efficiency, high yields, good quality of products, low costs, less waste and so on.
