Persimmon is the plant of Ebenaceae Diospyros . Persimmon Persimmon is the fresh or dried leaves, with good medical care and nutritional value. Persimmon is not only rich in Vc, flavonoids, amino acids and iron, zinc, calcium on the nutrients beneficial to human health, but also with antibacterial anti-inflammatory, antitussive bleeding, cancer prevention of cancer and other health care functions. Recent years, people have much attention on how to develop and take full use of persimmon leaves. Persimmon leaf tea was made from fresh leaves.In this study, Persimmon leaf tea flavonoids was extracted by ethanol and then separated and purified by AB-8 macroporous resin, we analyzed the structure of total flavonoids with LC-MS and relevant literature. In this study, Persimmon leaf flavonoids contained Iuteolin、Isoquercitrin、Quercetin、Kaempferol.Using DPPH method, xanthine oxidase, D-deoxyribose - Iron System Act, we compared and analyzed the free radical scavenging of flavonoids, the components purified by water, the components purified by 10% ethanol, the components purified by 20% ethanol, the components purified by 30% ethanol and the components purified by 50% ethanol. The results showed that persimmon leaf tea flavonoids, the components purified by water, the components purified by 10% ethanol, the components purified by 20% ethanol, the components purified by 30% ethanol and the components purified by 50% ethanol have different levels of free radical scavenging activity. The IC50 values of DPPH radical of flavonoids, the components purified by water, the components purified by 10% ethanol, the components purified by 20% ethanol, the components purified by 30% ethanol and the components purified by 50% ethanol were 0.011 mg/mL、0.10 mg/mL、0.035 mg/ml、0.017 mg/mL、0.020 mg/mL'0.036 mg/mL. The IC50 values of Superoxide anion radical of flavonoids, the components purified by 10% ethanol, the components purified by 20% ethanol, the components purified by 30% ethanol and the components purified by 50% ethanol were 3.03 mg/mL、19.93 mg/mL、17.49 mg/mL、7.41 mg/mL'2.98 mg/mL, and the maximum clearance rate of the components purified by water did not reach 50%. The IC50 values of hydroxyl radical of flavonoids, the components purified by water, the components purified by 10% ethanol, the components purified by 20% ethanol, the components purified by 30% ethanol and the components purified by 50% ethanol were 2.72 mg/mL、10.92 mg/mL、5.61 mg/mL、3.62 mg/mL、6.49 mg/mL'12.96 mg/mL. The results showed that PLF cleared Hydroxyl radical and DPPH free radical best,50% EPLF cleared O2-best.Using the model of carbon tetrachloride induceding acute liver injury in mice, we stutied the liver protection and antioxidant effects of persimmon leaf tea flavonoids and the purified components. The results showed that persimmon leaf tea flavonoids and the purified components on acute liver injury induced by carbon tetrachloride had different levels of protection. The components purified by water, the components purified by 10 % ethanol, the components purified by 20% ethanol, the components purified by 30 % ethanol and the components purified by 50% ethanol had inhibited the increase of ALT and AST activity, increased the content of serum albumin, increased SOD and CAT activity in damaged tissue of mice liver, reduced the contents of MDA and NO in liver damage tissue. It showed that 30 %EPLF-H was the best components to protect the liver in carbon tetrachloride liver injury model.
