Caenorhabditis elegans was introduced to construct type 2 diabetes mellitus (T2DM) model. Results showed that after synchronization of 48 hours, L1 formed dauers under a condition of 50 L1s per well on the microplate, regardless of enough food addtion on a level of 0.05%(w/v) E. coli OP50. It suggested that T2DM model was built successfully in C. elegans.Metformin and Na3VO4 were tested in this model. Na3VO4 has significant therapeutical effect on DR1564 T2DM model. L1s can bypass dauer to form adults by a ratio of 20.4%. Besides, the bypass effect is higher than the rescue effect. Meanwhile,22℃is the optimal temperature rather than 25℃for therapy. In contrast to Na3VO4, metformin has little therapeutical effect. This may be due to the differences on their mechanisms of action, suggesting that the model should be used to screen drug candidates targeted on downstream of insulin receptor.With regard to CB1370 T2DM model, both metformin and Na3VO4 has no therapeutical effect, it may be associated to its strong mutation on daf-2 gene.Prescriptions of traditional Chinese medicine (TCM) have distinctive advantage on the therapy of T2DM. Considering the speciality and the complexity of their mechanisms of action, the nematode T2DM model was investigated on screening traditional Chinese medicine. Four clinical prescriptions of TCM were chosen to test the efficiency of the nematode T2DM model. Results showed that both A decoctions and D decoctions have rescue dauers on a ratio of 28% in DR1564 T2DM model, Na3VO4 or metformin was not beneficial to the two TCM rescue effects. A decoctions and D decoctions have made Lls bypass dauers to form adults by a ratio of 40% and 45%, respectively, in DR1564 T2DM model when combined with Na3VO4. TCM showed more efficiency on T2DM model than Na3VO4, and this model was proved to be useful in screening TCM.In order to screen TCM efficiently and economically, TTC was investigated on the T2DM model. Results showed that the optimum condition is:39℃, TTC concentration of 3%, E. coli OP50 concentration of 0.0625% and treated for 120 mins, when C. elegans will intake TTC abundantly and efficiently. Besides, larva number had no effect on optical density, the number of C. elegans should be not less than 200 worms per well.
