Objective:The aim for the study is to investigate the inhibitory effect of 95% alcoholie extract of Rabdosia racemosa on tumor and its mechanism. Chronic myeloid leukemia K562 cells and its adriamycin-resistentvariant K562/ADR cells line were chosen as study objects to observe the effect of 2α,3α,23-trihydroxy-urs-12-en-28-oic acid on the cell proliferation.Methods:1. The method of experiment in vivo is performed to build a model of transplantable tumor in mice axilla by inoculating S180 sarcoma cell and H22 liver cancer cells subcutaneously in left axilla.60 mice inoculated with transplantable tumor were chosen and each of them was inoculated 0.2ml tumor cell.24 hours after inoculation, all the mice were divided into five groups randomly:model group, CTX positive group and alcoholie extract of Rabdosia racemosa low-dose group, middle-dose group, high-dose group. Each group has 12 mice,24 hours after inoculation they were treated with drugs for consecutively 10 days. Blood was collected and the mice were put to death in the 11th day, and then we weighted the tumor tissues of mice and calculated inhibitory rate.2. The effect on mouse immune functions:when the transplantation of S180 sarcoma cell and H22 liver cancer cells were executed, their thymus and spleen were culled and weighted for a further calculation of the thymus and spleen Index.3. Anti-oxidation effect inside the body: we use colorimetry to assayed the content of superoxide dismutase(SOD), catalase(CAT) and maleicdialdehyde(MDA) in mouse’s blood serum.4. The immunohistochemical method was used to measure the gene expression of Bcl-2, Bax and mutant P53 in tumor tissue.5. MTT assay was used to observe the effect of 2α,3α,23-trihydroxy-urs-12-en-28-oic acid on K562 and K562/ADR cells growth.Results:1. The alcoholie extract of Rabdosia racemosa has a significant compact of inhibition for the growth of transplanted S180 sarcoma and H22 liver cancer in mice. Inhibitory rate of low-dose group, middle-dose group and high-dose group were different from model group significantly(P<0.01 or P<0.05). The inhibitory rate of the high-dose group and middle-dose group transplanted S180 was more than 40%, alone with the inhibitory rate of thansplanted H22 was more than 30%.2. The alcoholie extract of Rabdosia racemosa increases the indexes of spleen. Moreover the low-dose group and middle-dose group were increased significantly(P<0.05). Contrarily, it has no influence on the indexes of thymus (P>0.05).3. The alcoholie extract of Rabdosia racemosa increases the activity of SOD and CAT and decreases the content of MDA. There was significant difference from the groups treated with drugs and model group(P<0.01 or P<0.05).4. The alcoholie extract of Rabdosia racemosa decreases Bcl-2 and mutant P53 protein expression, it also increases Bax protein expression in tumor tissue. The group treated with drugs were significantly different from that in model group (P<0.01 or P<0.05).5. Different concentrations of 2α,3α,23-trihydroxy-urs-12-en-28-oic acid inhibit K562 and K562/ADR cell proliferation significantly and in a dose-dependent manner.Conclusions:The alcoholie extract of Rabdosia racemosa can significantly inhibit the tumor growth. The related functional mechanism could be to improve the ability of anti-oxidation in the body. It also could be related to down-regulate the expression of the Bcl-2 gene, the mutant P53 gene and up-regulate the Bax gene expression.2α,3α,23-trihydroxy-urs-12-en-28-oic acid inhibit the K562 and K562/ADR cells growth in a dose-dependent manner.
