Hericum erinaceum, a kind of well-known fungi as food and medicine, has high nutritive and medicinal value. Besides polysaccharides and other macromolecules, Hericum erinaceum has some unique secondary metabolites. In this dissertation, the chemical constituents in the fruiting bodies of Hericum erinaceum were studied by using solvent extraction, column chromatography, thin layer chromatography, and some modern spectral analysis, the anti-tumor activities in vitro of some compounds were discussed, which enrich the study content of Hericum erinaceum and offer the academic foundation of its medical basis.After having extracted the dry powders of the Hericum erinaceum fruiting bodies with methanol at room temperature, the HE EtOAc and HE MeOH were obtained from the extractive by solvent partitions (ethyl acetate and then methanol), respectively. Each of the two parts were separated by column chromatography and purified by thin-layer chromatography, column chromatography and crystallization, respectively. Compounds 1 and 2 were obtained from HE MeOH, and compound 3 to compound 10 were obtained from the HE EtOAc. Then, the compounds were analyzed by nuclear magnetic resonance, mass spectrometry, UV scanners and infrared scanners. Along with reviewing pertinent literatures, structures of these compounds were identified by analysis datum.Compound 1 and compound 2 were D-erythritol and D-arabinitol alcohol respectively, while, compound 3 to compound 10 were 3β-hydroxy-ergosta-5,7,22- triene, 3β-hydroxy-5α,8α-epidioxyergosta-6,22-diene, stearic acid, oleic acid, hericene A, hericene D, hericenone C and hericenone D. Compound 8, named hericene D, was a new compound with side linoleic acid chain reported, which has the same mother nucleus structure as the hericenes A, B, C, i.e. 4-(3’,7’-dimethyl-2’,6’- octadienyl)-2-formyl-3-hydroxy-5- methoxybenzyl alcohol.In addition, the anti-tumor activities of compound 4, compounds 7 and 8, compounds 9 and 10 to the esophageal cancer EC-109 cells in vitro were studied. The results showed that at the concentration of 1×10-3mol/L, the inhibition rates of these compounds to the esophageal cancer EC-109 cells in vitro were all higher than 57%, in contrast to the one of 5-Fu, which was only 62.84% at the same concentration. The inhibition rates of these compounds were more than 91% higher than that of the positive control. And among, compound 4(3β-hydroxy-5α,8α-peroxide ergosta-6,22 -diene) showed more stable and higher inhibitive activity.
