The avian flu was one of the deadly infectious disease which caused by an Avian Influenza Virus, and spreaded in the Avian species. In this study, The recombination Avian Influenza Virus (H5N1 hypotype, Re-5 strain)was concentrated by the concentarion technology of the tangential flow ultra to filter, By selected the 50,000, 100,000 and 300,000 different aperture ultra filter diaphragms, we established the ultra has filtered the system, the antigen fluid of the recombination Avian Influenza Virus was to be concentrated in this study. This experiment established by the Millpore medium box type antigen, respectively selected the interception molecular weight is, has conducted the concentration experimental study to the bird flu virus antigen fluid. The result demonstrated that the the antigen fluid of the recombination Avian Influenza Virus could be concentated very greatly by the ultra filter diaphragms of the different molecular weights, and the returns ration upto 100% ,compared to the same period, the titre enhanced. The vaccine was pruducted, After the concentration antigen emulsification compounds, the chichen were immuitied, in the 10, 14, 21, 40d, the immunity resistant determination result surpassed the vaccine made by the conventional antigen.The rule of the immune body increased and decreased indicaded that the immune body produced stably by the concentration vaccine and the immune response process had met the oil emulsion vaccine requirements; The immunity resistant duration was long; The immunity side reaction was smaller than the conventional vaccine by far. Concentrated the antigen to the Avian Influenza Virus to carry on the different concentration multiple, the antigen to make in the vaccine immunity resistant determination time to be able to achieve the immunity effect and the immune body eliminates the rule the research to discover, the immune body could create by different group vaccine, but the titre of the immune body was concerned with the concentration multiple of vaccine in 10th,the multiple bigger immune body is higher,the group of the 3-5 times of concentration antigen could producted earlier and higher between 5-10d, the titer could achieve 10; along with the antigen concentration multiple, the immune body increased between the groups, and the immune body homogeneity increases along with the concentration multiple was more homogeneous in the group. The anbody immunitied by each concentration group antigen vaccine were higher than the the conventional group antigen vaccine immuntied. After each group of antigen system seedlied, the rule of the immunity increased and decreased was not demonatrated by the vaccine antigen content and the different immunized doses.the inhibitory action of the prducted antibody which producted by the concentration antigen’s content may be studied further.From the each group of the concentration vaccine, the side reaction was existed, but the degrees were differented. under the same condition, the side reaction of the concentrated vaccine was smaller than the conventional vaccine, the consequence was possibly caused that some inorganic salt, the small molecular protein, the endotoxin elimination result in the concentration process, the adjuvant side reaction was bigger than the concentration antigen or not, the further experimental study and the quantification of was to be carry out.
