Epidermal cells represented the accessible model systems for studying cell pattern and specification in plant.There were little reports about the development process of long cells, short cells, prickle hairs and subsidiary cells from single and double stomatal rows in rice (Oryza sative) leaves. In this paper, we studied the morphogenesis of these cells by the dental resin impression method, showing the development process of subsidiary cells in single and double stomatal rows in detail and summarizing the morphogenesis pattern of long cells, phellem cells, silica cells and prickle hairs. Long cell in stomatal row and guard mother cell (GMC) came from the same precursor cell and subsidiary cell was produced by the long cell flanking the stomatal row. Normal long cell and phellem cell were starting at the different cells in the base of immature rice leaf, and prickle cells came from the cell protuberance on the phellem cell row and silica cell originated in two phellem cell.In an epidermal mutant 245 epidermal mutant (245em) in rice, the serrated lobes of leaf long cells were smoother than wild type and the phenotype did not weaken or vanish. The lobes of mesophyll cells were also gentle in 245em leaves, and the change was lessening with the leaf age increasing. However, the mutant phenotype did not disappear. The silicon content in 245em leaves was about half of ZH 11 leaves. The experiments of drought and 20% PEG4000 treatment indicated that 245em was sensitive to drought. In the water loss experiments of 3rd and 4th leaves, the water loss rate in 245em leaves was higher than ZH 11.After treating with 100 mM NaCl, 245em withered faster than ZH 11 and the survival rate in 245em was lower than ZH 11. Silicon could helpe plants to overcome water and various mineral stresses. Therefore, the reduction of silicon content decreased the resistant of 245em to abiotic stresses.245em was fertile, and its mutational phenotype could pass down. Southern blotting indicated that 245em might contain single copy insertion of T-DNA.
