The animal disease,which is one of the restrictive factors on animal husbandry development,cause a huge economic losses of animal production.What is more it is harmful to human health.The disease resistance breeding is one of good solution which aims to elevates the immunity of animal and animal antidisease aspects.Five resistance candidate genes(FUT1,HLA-B2,MX1,CXCL12,EHMT2) were selected to look for single nucleotide polymorphisms(SNPs) and detected polymorphisms in eleven breeds and associate with immune traits,such as Hog Cholera antibody level and blood routine examinations.We also study on the genotypic relative risk in health and infected pigs.The major results were described as follows:1.6 SNPs were found in different breeds.The methods used to detect the 6 SNPs was established and the 4 of them(HLA-B2,Mx1,CXCL12,EHMT2) was detected by introducing restriction enzyme sites.PCR-BclI-RFLP was developed to detect 79 A/G substitution at the intron 25 of HLA-B2.PCR-FnuDII-RFLP was developed to detect 140 C/T substitution at the exon 13 of MX.PCR-PvuⅡ-RFLP was developed to detect 262 C/T substitution at the intron 27 of EHMT2.3 mutations in CXCL12 gene were found in the different breeds. In addition,genotype protocols for 7462 C/G at the intron 3 resulting in BsmAI polymorphism,7331 A/G at the intron 3 resulting in BSTN1 polymorphism,and 7373-7374 AC/CT at the intron 3 resulting in Bsrl polymorphism were established in this study.2.7 SNPs were analysesed in ten breeds or swinery.Three genotypes were detected in the exotic breeds,but only GG genotype was detected in Chinese breeds and Asian Wild boar at the FUT1 polymorphism.No AA genotype was detected in Duroc,Large White,Qingping,Jianli,While only GG genotype was detected in Small Meishan,and three genotypes were detected in other breeds at the HLA-B2-BclI-RFLP polymorphism.No TT genotype was detected in Duroc,Large White,Small Meishan,Qingping,Jianli,Jinhua and Wild boar,and three genotypes were detected in others breeds at the MX1-FnuDⅡ-RFLP polymorphism.No TT genotype was detected in Duroc and Jianli,While only CC genotype was detected in Qingping,and three genotypes were detected in other breeds at the EHMT2-PvuⅡ-RFLP polymorphism.No CC genotype was detected in Duroc, and three genotypes were detected in others breeds at the CXCL12-BsmAI-RFLP polymorphism.Only GG genotype was detected in Qingping and Jianli,While no AA genotype was detected in Wild boar,and three genotypes were detected in other breeds at the CXCL12-BSTN1-RFLP and CXCL12-Bsr1-RFLP polymorphism.3.FUT1 is a candidate gene of Escherichia coli receptor,which was mutated in 307.Three genotypes were detected in the exotic breeds,but only GG genotype was detected in Chinese breeds and Asian Wild boar.The relative risk associated with FUT1 gene polymorphism in the four clinical types of healthy,recovery,during sickness and death pigs after infected.The relative risk of GG genotype may be 2.040 times compared with AA genotype(P=0.025).4.The relative risk associated with HLA-B2 gene in intron 25 polymorphism in the four clinical types of healthy,recovery,during sickness and death pigs after infected.The relative risk of AG genotype may be 0.540 times compared with AA genotype(P=0.0504).The relative risk associated with the polymorphism of HLA-B2 gene in the healthy and disease(including recovery,during sickness and death pigs) after infected.The relative risk of GG genotype may be 0.513 times compared with AA genotype(P=0.0463).Genotype GG in hemoglobin content and Hematocrit at 0 day old in Landrace was more than Genotype CC and CG(P<0.05).Pseudorabies virus antibody level(AA>GG>AG) in Duroc×Landrace×Yorkshire at the HLA-B2-BclI-RFLP polymorphism was significant(P<0.05).5.The relative risk associated with Mx1 gene in exon 13 polymorphism in the four clinical types of healthy,recovery,during sickness and death pigs after infected. The relative risk of CT genotype may be 2.041 times compared with CC genotype (P=0.0019).The relative risk associated with Mxl gene polymorphism in the healthy and disease(including recovery,during sickness and death pigs ) after infected.The relative risk of CT genotype may be 1.936 times compared with CC genotype (P=0.0056).The difference for Pseudorabies virus antibody level(CT>CC>TT) at 0 day old in the Landrace at the Mx1-FnuDⅡ-RFLP polymorphism was extremely significant(P<0.01).The difference for Porcine Reproductive Respiratory Syndrome virus antibody level(TT>CC>CT) at 0 day old in the Landrace at the Mx1-FnuDⅡ-RFLP polymorphism was significant(P<0.05).The difference for erythrocyte counts,hemoglobin content and Hematocrit(CC>CT>TT) at 17 days old in the Landrace at the Mx1-FnuDⅡ-RFLP polymorphism was significant(P<0.05).6.The relative risk associated with CXCL12 gene at CXCL12-BsmAI-RFLP polymorphism in the four clinical types of healthy,recovery,during sickness and death pigs after infected.The relative risk of CG genotype may be 0.652 times compared with CC genotype(P=0.0235).The difference for mean corpuscular volume (GG>CC>CG) at 0 day old and Pseudorabies virus antibody level(CG>CC=GG)at 17 days old in the Landrace the CXCL12-BsmAI-RFLP polymorphism was significant (P<0.05).The difference for mean cell hemoglobin concentration(GG>AG>AA) at 0 day old and leukocyte counts(AA>AG>GG) at 32 days old in the Landrace at the CXCL12-BSTN1-RFLP polymorphism was significant(P<0.05).The difference for platelet distribution width,platelet volume and platelet large cells ratio(AA>GG>AG) at 17 days old in the Landrace at the CXCL12-BSTN1-RFLP polymorphism was extremely significant(P<0.01).7.The relative risk associated with EHMT2 gene at EHMT2-PvuⅡ-RFLP polymorphism in the four clinical types of healthy,recovery,during sickness and death pigs after infected.The relative risk of GG genotype may be2.414 times compared with TT genotype(P=0.0061).The relative risk associated with EHMT2 gene in the healthy and disease(including recovery,during sickness and death pigs) after infected.The relative risk of GG genotype may be 2.411 times compared with TT genotype(P=0.0031).The difference for Hog Cholera antibody level and Pseudorabies virus antibody level(TT>GG>GT) in Duroc×Landrace×Yorkshire at the EHMT2-PvuⅡ-RFLP polymorphism was extremely significant(P<0.01).
