Phosphoenolpyruvate carboxylase (pepc) is a kind of catalyzing enzyme,which can catalyze CO2 fixation initial reaction in C4 plant dicarboxylic acids recycling ways,making the C4 plants’photosynthetic rate obviously higher than the C3 plants’, especially under the condition of strong light and temperature.Zeamays C4 type PEPCase gene recently was successfully transformed into C3 crop rice , provideing the excellent prospective to genetic engineering technology that transforming the C4 type PEPCase gene into the C3 crop to improve photosynthetic efficiency. This technology had becamed the important breeding direction at home and abroad.Sugarcane is one of the highest photosynthetic efficiency crop. Plant expression vectors pC1380PEPC containing complete sugarcane pepc gene with primal promoter was transformed into indian super restorer lines rice“hangl”calli by Agrobacterium tumefaciens, and we expected significantly increasing hang1’s photosynthetic efficiency and production. Meanwhile , we analyzed genetic characters of transgenic restorer lines rice N175.The results were as follows:1. The pC1380PEPC containing complete sugarcane pepc gene with primal promoter was transformed into hangl calli by Agrobacterium tumefaciens, and 172 regenerated rice plants were got.Results of PCR detection showed that the exogenesis pepc gene can be detected in 112 regenerated plants, and there were no positive results in control plants.2. PEPC activities of the leaves from the transgenic restorer lines rice N175 were measured. The result demonstrated that most of photosynthesis of the transge- nic rice expressed significantly higher than those of the non-transformants.3. Results of southern blot indicated that the sugarcane pepc gene had been successfully transformed into the transgenic restorer lines rice N175.
