Dendrobium nobile Lindl.,one of the rare Chinese medicinal herbs,grows in the Sichuan,Guangxi,Yunnan,Guizhou province in China.Pharmacological studies have demonstrated that D.nobile Lindl.,and its constitutes have relieve fever,depress blood pressure and anti-inflammatory.This plant has many specific charatcistics,such as the complex mechanism of the growth process,slow growth rate,hard living environment, extremely low growth and development rate and so on.Because of these biocharacteristics and the uncontrolled collection,the wild resources of Cistanche deserticola in China is on the edge of extinction.In order to protect this rare plant resourses and for better exploitation,on the basis of reviewing the recent study on D.nobile Lindl.,we induced PLBs from the roots of D.nobile using plant tissue culture techniques.The best growth medium condition was as follows:1/2MS+YE500mg/L+NAA 1.0mg/L+6-BA 2.0mg/L+sucrose 15g/L+agar 4.5g/L,and light time 12h/d,light intensity 100μmol.m-2.s-1was propitious to the PLBs grow.We add yeast extractive,casein hydrolysate and potato extractive with different concentration to the medium to research the effects on the PLBs growth and polysaccharides yield.the experiment show that the yeast extractive with 500mg/L can enhancement polysaccharides yield most.It’s 1.75 times to the control.In the experiments,the effects of pH and the illumination were investigated.We found that the polysaccharides production is higher when pH is 5.8,and light time 12h/d and light intensity 100μmol.m-2.s-1was propitious to produce polysaccharides.The plant differentiate from the D.nobile Lindl.PLBs have been implement with this experiment.The Research on the influence to regeneration plant of D.nobile Lindl.by the phylohormone instruct that by adding 6-BA 2.0mg/L and KT 0.5mg/L can induce the D. nobile Lindl.PLBs differentiate to regeneration plant. The effect of phylohormone were investigated in the course of regeneration plant cultivation.the result showed that the suitable medium for the regeneration plant cultivation is as follow:1/2MS+NAA 0.5mg/ml+6-BA 2.0mg/ml sucrose 30g/L+agar4.5g/L+3mg/LPP333.In this research,we have established a method for determination of polysaccharides to control the quality of the PLBs and the cultured plant of D.nobile Lindl.
