The mainstay of treatment for fracture is surgery. But with the development of medical molecular biology,scholars have found many growth factors,which play extremely important role in the process of fracture healing. As a start of osteogenesis factor,BMP can induce ectomesenchymal cells differentiation into bone cells.However rh BMP-2 has not been reported on clinic of veterinary medicine. As a kind of Mitogen-activated,b FGF has a good effect on promoting the division of mesenchymal cells and bone forming cells and angiogenesis,and promoting the form action of newbone and acceleratethe bone healing. Tobramycin as a kind of higher antimicrobial sensitivity antibiotics,can be effective treatment of the infection in the process of fracture healing.So,in this experiment,due to its low immunogenicity,slower degradation rate and a special three-dimensional network structure,FG was used as a slow release carrier which was mixed with rh BMP-2,b FGF and tobramycin in canine tibial fracture model. The purpose is to explore the effects of FG recombining composite rh BMP-2,b FGF and tobramycin on pets fracture healing and provide a new treatment for the fracture treatment of pet dogs.The experiment selected 12 small healthy dog with similar age and size,making completely defect transverse fracture model at both sides of each dog tibial. Each right tibia fracture model applies with FG composite of rh BMP-2,with b FGF and tobramycin as experimental group,and lift tibia fracture model should not be used any drugs as control group. The author observed and recorded the disappearing time of postoperative red,swelling,fever. Then the author did X-ray observation after 2weeks, 4 weeks,8 weeks,12 weeks and 16 weeks,and evaluate each fracture time point according to the standard of X image reading.Take advantage of RA measured the BMD in the healing area postoperative 4 weeks,8 weeks,12 weeks and 16 weeks.Tahe author was used vernier caliper to measure fractures callus volume after 4 weeks and 8 weeks. the bone callus volume determination. After 16 weeks,the bilateral tibia will be removed and it will be placed on the universal tensile testing machine for three point failure test,which measures the maximum load and the maximum displacement.At postoperative,the disappearing time of red,swelling and heat in experimental group is 6.61 ± 0.93 days,which is shorter than that of the control group 9.33± 0.94 daysand the difference is significant(P<0.05).Through X-ray observation that a small amount of callus appeared at 2 weeks post-operative.Generating the cortical bone and Bone marrow cavity healed well postoperative 12 weeks can be used as an the standard of clinical healing,but bone marrow cavity healed bad in control group at 16 weeks post-operative.The grading result of X-ray of experimental group is significantly higher than the control group after 2 weeks,4 weeks,8 weeks,12weeks(P<0.01),while the score of experimental group 30.00±0.00 is not significantly andcontrol group 28.00±3.09 at week 16 post-operative,and experimental group29.33±2.00 after 12 weeks is not significant than experimental group 30.00±0.00 after16 weeks(P>0.05). The BMD in experimental group after 4 weeks,8 weeks,12 weeks,16 weeks are all higher than those of the control group,while comparing the deference between the BMD 1.63±0.42g/cm3 in experimental group and the BMD 1.60±0.33g/cm3 after 12 weeks,the result is not significant(P > 0.05).The bone callus volume2.54±0.11cm3 after 4 weeks and 1.88±0.05cm3 after 8 weeks are all significantly higher than those in control group 1.54±0.14cm3 and 1.42±0.22cm3(P < 0.05). At 16 weeks post-operation,measuring tibia under the maximum load and the maximum displacement when bending occurs on three failure experiment,the experimental group 72.11 ± 2.00 N and 1.20 ±0.05 mm are all significantly higher than those in the control group of 6.45 ±2.51 N and 1.00 ± 0.04mm(P<0.05).Experimental results show that FG mixed with rh BMP-2,basic b FGF and obramycincan speed up fracture healing,shorten the fracture healing time,promoted fracture callus generated in the process of bone mineral deposits,and bone biomechanical strength of the recover.
