Using 19 pairs of microsatellite primer to detective the genetic background of the Specific Pathogen Free pedigree, one hundred pigs, Yorkshire and Landrace which imported from Canada by Harbin Veterinary Research Institute, Chinese Academy of Agricultural Sciences. They are SPF pigs without Swine Fever, Porcine Reproductive and Respiratory Ryndrome, Swine Flu virus or pathogen. And then Yorkshire×Landrace and Landrace×Yorkshire crossbreeding pigs are also been detectived. Finally, we using the low resolution PCR-SSP technique selected the SLA haplotype of Large White and Landrace individuals.Discussed the feasibility of the SPF pigs as an experimental animal model. The results are as follows:1. All of the 19 microsatellite loci by Canadian SPF Large White and Landrace pigs are highly polymorphic, especially S0155, S0143, S0178, Sw857 and Sw936 sites have a significant distributed in the two herds, which of them are able to a candidate site of species identification.2. The average heterozygosity of the two groups between Large White and Landrace in 19 microsatellite loci was 0.6066 ' 0.5877(between 0.5 and 0.7). The observed heterozygosity(Ho) and expected heterozygosity(He) was 0.5875、0.5877 and 0.6156、0.6066 has no discrepancy(P>0.05). All of the two herds meet the conform of closed colony in genetic characteristics, which of them can be used as an experimental animal model make use of swine disease research and vaccine in swine.3. The average heterozygosity of Yorkshire×Landrace crossbreeding pigs is 0.7322 greater than 0.7, it is not in line with closed colony herd. All the 19 microsatellite loci were negative but IGF1 and S0218 in genotype differentitation of F-stastic(Fit),which showed the heterozygosity are excess need further breeding.4. Results of the structure and genetic differentiation showed that the differentiation within the group of Landrace and Yorkshire were small and genetic structure were stability.5. We filtered eight SLA-1 BLANK alleles at the sample of 2922, 2992, 2956, 2952, 3033, 3065, 5131 and 5132.
