In this study, two genes(growth hormone(GH) gene and myostatin(MSTN) gene)related togrowth of Silurus lanzhouensis were seclected to study the detection of SNPs and polymorphism analysis. As experimental material, 100 Silurus lanzhouensis came from the same family and the same collection pool.With the methods of sequencing and single-strand conformation polymorphism(SSCP), SNPs of GH and MSTN gene were detected and molecular markersrelated to growth traits were found,which benefit for the protection and breeding of Silurus lanzhouensis resources. Growth hormone(GH) genes containing a complete open reading frame were cloned from genomic DNA of Silurus lanzhouensis using long PCR and T-A cloning techniques for the first time, and the accession numbers in Gen Bank is KM215221. At the same time, these sequence were analyzed by DNAstar and other bioinformatics software. The full-length of S. lanzhouensis GH gene is 2 067 bp, including five exons and four introns, and the complete coding sequence(CDS) is 603 bp. The size of 5 exon size were 10 bp, 140 bp, 117 bp, 132 bp and 204 bp respectively and 4 intron were 229 bp, 103 bp, 565 bp and 103 bp respectively.The junction sequence of introns and exons of S. lanzhouensis GH gene was in accordance with the rules of gene composition.The coding region encoded a polypeptide with 200 amino acid residues.Based on the amplification and SSCP analysis and sequencing of Silurus lanzhouensis GH gene, the results showed that the sequence of the 1, 2, 4, 5 exon and the 3, 4intron were quite conservative, and only 1 SNPs were found in 3 exon. SAS software was used to analyze growth traits(including weight, body length, body height and the length) of different genotypes of Silurus lanzhouensis group.The results indicated that the mutation has significant effect on body weight and body length(P<0.05). The weight and longs of AA homozygous individual was significantly higher than BB individuals(P<0.05); The weight of AB individual was significantly higher than BB mutation.MSTN gene:Primers were designed by using MSTN gene sequence of Silurus lanzhouensis related species.polymorphism analysis of 3 exonsin MSTN gene of Silurus lanzhouensis was performed and found A/G single-base mutation in the first exon.Through the analysis, the results showed that different genotypes of MSTN gene have significant effects on body weight and had no significant effect on body length of growth properties. The weight of CC genotype and CD genotype were significantly higher than other individuals.By Screening the GH and MSTN gene in Silurus lanzhouensis, two single nucleotide polymorphisms marker was found and their relationship with growth related traits of Silurus lanzhouensis was analyized.The results showed that these two genes were the main effect genes affected the development and growth of Silurus lanzhouensis.G/A mutation in the third exon of GH gene and A/G mutation of MSTN gene can be developed as molecular markers for growth character selection of Silurus lanzhouensis, also as theoretical basis for the protection and breeding of Silurus lanzhouensis.
