In recent years, the technology of transplanting in well-cellar has greatly changed the industrial structure of tobacco in the southwest area of our country. Tobacco seedlings are no longer required to cultivate 7 to 8 leaves but only about 5 leaves, 3-4 cm plant height and 1.5 cm stem girth.The technology greatly shortens the cycle of seedling, furmore promote the research and development of cultivation technology of tobacco seedling. In order to cultivate suitable tobacco seedling and explore the appropriate measures and technology to cultivate transplanting seedlings in well-cellar, this study analyses the effect of new-style artificial environment seedling room, fertilization, germinating trays, copper sulfate and rooting powder to the growth and development of Yunyan 87 tobacco seedling. The results are showed below.1. Using artificial environment seedling room and conventional floating seedling shed to cultivate tobacco seedling, we found that artificial environment seedling room had many advantage, such as smaller area and shorter seedling period. Automation control system ensured that the daily accumulated temperature of nursery room was 2.9 times higher than conventional greenhouses, shortened the seedling growth period around 23 d, and reduced the cost of 50%. But the tobacco seedling cultivated in the artificial environment seedling room was weak and extended the period of rooting after transplanting. Later we need to improve the management measures to enhance the adaptability and resistance of seedling root.2. The three levels of rooting powder were used to fill the root and spray on the leaf. The study found that the growth of the root system was inhibited by rooting powder filled root, while rooting powder sprayed on the leaf promoted the growth of the root system. When we sprayed 20mg/L rooting powder on the leaf, the growth of tobacco seedling was the best.3. By using 200, 308, 570 three kinds of germinating trays, the study found that the longitudinal growth and the number of root under trays were increased. But with the trays hole increased, the root appeared weaker, and the period of rooting was longer after transplanting. By using 200 germinating trays the lateral growth of root and root activity was higher, plant height and stem was appropriate, the period of rooting after transplanting was shorter, root activity and dry weight of seedling was the highest.4. We applyed two kinds of NPK ratio(10:8:10, 10:10:20) to tobacco seedling ratio and four kinds of NPK content(nitrogen content per plant was 9, 11, 13 and 15 mg). The study found that high proportion of P and K promoted the increase of plant height, stem circumference, leaf number, leaf area, soluble protein and free amino acid content of tobacco seedling. But by exerting high content of fertilizer, the root growth of tobacco seedling was restrained. In the treatment of N:P:K=10:10:20, nitrogen content per plant 9 mg, the growth of tobacco seedling performed the much best.5. By applying 6 kinds strength of copper sulfate solution in the seedling pool, we found when the copper sulfate concentration was higher than or equal to 50 mg/L, the area of algae was less than 8% in the seedling pool. The growth of root was restrained, of which root length, root shadow area, root surface area, root number, root scan index and root activity was low. Applying 100 mg/L copper sulfate solution, the growth of root was best and more biomass accumulation above ground of tobacco seedling.In order to cultivate suitable tobacco seedling and explore the appropriate measures and technology to cultivate transplanting seedlings in well-cellar, We can use artificial environment seedling room and improve the management measures to enhance the adaptability and resistance of seedling root. We spray 20mg/L rooting powder on the leaf to promot the growth of the root system.Using 200 germinating trays to cultivate tobacco seedling. We apply N:P:K=10:10:20and nitrogen content per plant 9 mg to promot the growth of tobacco seedling. Applying 100 mg/L copper sulfate solution to reduce the area of algae.
