Raccoon aquaculture is big in our country,but not a raccoon powers, particularly in the field of raccoon breeding,feeding,reproduction and other developed countries there is a gap. Raccoon is a kind of important economic animal, raccoon skin is one of the important raw material production of fur. Raccoon coat colors are important factors affect the value of a raccoon skin,and the formation of the colour is considered to be the body to produce melanin. In vertebrates,according to a study by genetic mitf transcription factors associated with small eye malformation is the most important transcription factors affect the colour. At home and abroad about raccoon coat genetic and study the formation mechanism of less.This experiment selected wusuli raccoon,JiLin white raccoon,black raccoon, reddishbrown mutation raccoon raccoon of four different colour as the research object. Raccoon are studied by using PCR and direct sequencing method part of MITF gene polymorphisms of exon and intron 1 correlation with the colour,and to obtain mitf sequence of bioinformatics analysis.Purpose:through the wusuli raccoon,JiLinBai raccoon,black raccoon,reddish-brown mutation raccoon four different colors small raccoon eyes deformity associated mitf transcription factor gene cloning,identification,and carries on the bioinformatics analysis. And part of mitf gene exon and intron 1 of SNPs detection, to study the mitf correlation with four different colors raccoon. To clarify raccoon coat mutant form of the study of molecular mechanisms.Methods:using PCR amplification wusuli raccoon mitf genes,and bioinformatics analysis. And direct sequencing method for four kinds of color, a total of 120 individual SNPS detection.Results:the first successful wusuli raccoon mitf length gene sequences; Bioinformatics analysis showed that mitf gene A(404) : 32.1%; T(262) : 20.8%; C(328) : 26.0%; G(266) : 21.1%,characterized by A > C > T > G, among them A + T(52.9%) than C + G(47.1%). mitf theory of protein molecular weight is 46908, isoelectric point(PI) was 5.84,no stability coefficient is 64.34,the total average hydrophilic coefficient is 0.640,located in the nucleus. In raccoon measured SNPs mitf genes inside and outside show 9 screening to 2 SNPs, screening in the introns 1 to 1 TAC loss of mutations. They are,respectively,g. 132 C> T, g. 237 G > A and g. 106 delTAC.Conclusion: the cloned from wusuli raccoon mitf gene localization in the nucleus, the protein as unstable acidic protein hydrophobicity,exon region,did not find effective mutations mitf not four raccoon coat mutations of main effect genes, or speculating on its genetic stability is better, for the next step ideas,the color of the raccoon key genes and provide experimental basis.
