Maize is an important grain, and one of the important industrial crops to feed,industrial and new energy also other many uses, in terms of planting area and planting scope is the largest crop species in China, with the number of new varieties of maize increased year by year and the development of seed industry at the same time,varieties of authenticity identification requirements also increased dramatically.Traditional field planting appraisal due to the growing cycle is long and vulnerable to the impact of the surrounding environment, which restricts the development and application of large scale.DNA fingerprint technology is not influenced by the environment, the advantages of rapid, accurate especially in molecular markers with codominant of SSR(Simple Sequence Repeats) and SNP(Single Nucleotide Polymorphisms), reflect the specific fragment or Sequence information marked by UPOV- BMT molecular test guidelines recommend as a symbol of the optimization of varieties authenticity identification and database building.SNP has the following advantages compared with SSR become varieties authenticity identification and database to build the most development potential of markers: the first one is the distribution of density in the genome in higher and more evenly distributed; secondly is easier to achieve platform data integration and comparison. Finally, it has a higher flux.In the experiment one, the capsule DNA was analyzed by using the SNP Golden Gate genotyping method to obtained the data. The experiment use two sets of parents/F1 samples as materials, contain Jingke 968, Jing 724, Jing 92, Zhengdan 958, Zheng58 and Chang 7-2. Through the analysis of the obtained rate, signal value and typing effect from the seed capsule DNA data. We successfully establishment a suitable set of SNP chip for the classification of maize capsule tissue DNA extraction method.Experiment two through the analysis of 96 representative varieties of maize DNA data acquisition rate, signal value, typing results to explore the SNP standard fingerprint data construction technology and ideas. The experiment two establish the maize variety released primary SNP fingerprint database, and fingerprint screening and SNP database based on the construction of an important work, namely authenticity identification and purity detection of core sites screening lay the foundation.
