Study On The Gemmae Propagation And Histochemical Localization Of Alkaloids Of Huperzia Serrata And Fresh Materials Extract

Huperzia is a traditional Chinese herbal medicine, medicinal history dates back thousands of years. Chinese scientists in the early 80 ’s and extraction were given huperzine A. Huperzine A is a kind of alkaloid and is a high efficiency, low toxicity, highly selective as an effective natural ACh E inhibitors, which used to treat senile dementia, myasthenia gravis, memory recession and other diseases.In the paper five parts experiments were processed as follows:(1) To explore the factors influencing the gemmae germination of Huperzia serrata and growth condition. The effect of culture medium, culture temperature,and light intensity and hormone on the gemmae germination condition of Huperzia were studied,and the growth morphogenesis during the course of gemmae germination of Huperzia serrata was observed at the same time.The best culture temperature for the gemmae germination was(22±2℃),and the germination rate of the optimum medium for Moore,the germination rate was as high as 82%,and the average plant length could be up to 14.6mm. The second culture medium for the gemmae germination was Knudson C,and the germination rate was 64%,the average radicle length is about 5.2mm. Darkness is conducive to gemmae germination,and the germination rate was 92%,the average radicle length is about 9.1mm. Under 800 lx weak light,the radicle grow slowly,and the radicle length could be up to 8.7mm. 1600 lx light irradiation exhibited some inhibition for the root growth. And the hormone for gemmae germination and growth exhibited some inhibition.(2)In different age stages of order of huperzine A contents in xiangxi plots was seedlings>adult plants>young plants.in the root,stem and leaves in different organs of huperzine A have some distribution of,leaf>stem>roots.(3)Study on the alkaloids of huperzia in all morphological distribution of whole herb,fresh materials of tissue reaction showed that alkaloids of huperzia is mainly distributed in the stem column of phloem and cortical cells;focus of leaves surrounding the stomata cells;looming red-orange particles dispersed in the root cortex,and epidermal cells in the column.(4) Get a best live release conditions of huperzia serrata extract huperzine:0.75% tartaric acid as extraction solvent with a solid-to-solvent ratio of 1:36(g/m L) and an extraction time of 43 min,the yield of huperzine A was 8.55×10-7g,and close to the forecast values,the Show method.
