A Study On The Biological Characteristics Of Wild Agaricus Bitorquis Qinghai Plateau

Agaricus bitorquis belongs to Eumycota, Basidiomycotina, Hymenomycete,Homobasidiomycetidae, Agaricales, Agaricaceae, Agaricus. Agaricus bitorquis is a new mushroom development with high value and biological activity. This article we choose Agaricus bitorquis from Qinghai Province to research. Its liquid fermentation nutrient conditions, fermentation technology was studied. The results are as follows:1. Study on the condition of Agaricus bitorquis solid plate cultureThrough single factor and orthogonal experiments, we chose a better solid plate culture medium for Agaricus bitorguis,that is medium formula:maltose 30.0g,KNO3 8.0g, KH2PO4 0.5g, Mg SO4 0.5g, agar 18.0g, H2 O 1000mL. The daily growth rate reaches 9.4mm/d.2. Study on nutritional conditions and fermentation process of Agaricus bitorquis liquid fermentationThrough single factor and orthogonal experiments, we chose a better fermentation process for mycelial biomass, that is nutritional conditions of maltose30.0g, peptone 5.0g, KH2PO4 0.5g, Mg SO4 0.25 g, vitamin 15.0mg, H2 O 1000mL;Fermentation process conditions of 140 mL/250 mL liquid volume, 25℃, 210r/min,5% Inoculum, 5 glass beads, p H value of 6, culture 6 days, this process we got relatively high biomass, reach to 1.3g/100 mL.Through experiment, we chose a better fermentation process for extracellular polysaccharide production, that is nutritional conditions of maltose 30.0g, peptone5.0g, KH2PO4 0.5g, Mg SO4 0.25 g, vitamin 15.0mg, H2 O 1000mL; Fermentation process conditions of 140 mL/250 mL liquid volume, 25℃, 210r/min, 5% Inoculum,5 glass beads, p H value of 6, culture 6 days, this process we got relatively high extracellular polysaccharide production, reach to 0.80g/100 mL.3. Study on the culture conditions of Agaricus bitorquis two species of bacteriaThrough experiments, the optimum conditions for the culture conditions of Agaricus bitorquis two species of bacteria are determined:Cottonseed shell medium: Wheat 70%, cottonseed shell 30%, KH2PO4 0.5%, Mg SO40.25%, glucose 3.0%;Sawdust Medium: wheat 90%, sawdust 10%,KH2PO4 0.5%, Mg SO4 0.25%, glucose3.0%;Wheat straw medium: Wheat 80%, wheat straw 20%, KH2PO4 0.5%, Mg SO4 0.25%,glucose 3.0%.4. Study on the culture conditions of Agaricus bitorquis solid substrateIn the experiment, the optimum Agaricus bitorquis solid matrix culture condition was determined. The wheat was solid matrix and the nutrient solution was glucose 20.0g, beef extract 5.0g, Mg SO4 0.5g, KH2PO4 1.0g, VB110.0mg, H2O1000 mL.
