The experiment based on the separated ORFV proliferation culture in lamb Testicular cells, continuous inoculation weaken Virulence, Detect pathogencity of 2month lamb, select different generation of cell venom by TCID50 and virus titer was tested, draw out cell venom from 20th generation、 50thgeneration、70thgeneration、90thgeneration determination of TCID50 and measure virus content by fluorescence quantitative PCR method at the same time。The separated ORFV proliferation culture in lamb testicular cells, inactivated by formaldehyde for 48h at 37℃, Use 37℃ Orf Virus for 48h, Use maintained ISA 201 VG adjuvant and Montanide ISA 11 R VG adjuvant prepare WOW inactivated vaccine, And check the appearance, stability and viscosity of the vaccine., then select 2month lamb do the safety inspection and immunity test。The test result shows that, while Orf virus inoculation in lamb testicle cell to 90generation, virus virulence will markedly diminished, without causing a morbidity of lambs, TCID50 detection of virus 20,50,70,90 generation shows virus titer is between 10-5.5~10-6.6。 The preparation of ORF inactivated vaccine bacterium check tested and proved to be qualified, Appearance, stability and viscosity are conform to quality standards, Vaccine safety test shows, lamb is not morbiditied, antibody titers is between 1:2-1:8 In Immune efficacy test, In animal challenge protection test201 Adjuvant Inactivated Vaccine lamb and 11R Adjuvant Inactivated Vaccine lamb have certain protective.
