Transmissible gastroenteritis (TGE),is an acute and highly contagious disease characterized by vomit and severe diarrhea, and induced by transmissible gastroenteritis virus (TGEV).It is very important to develop a kind of simple and high accurate detection method on the prevention and control of the disease. In this study, a pair of specific primer was designed and synthesized according to the sequence of spike (S) protein gene of TGEV published on GenBank. According to the sensitivity and specificity test, The results showed that the established RT-PCR method can be used in the detection of TGEV.204 diarrhea fecal samples were collected from seven cities of Inner Mongolia during 2015 to 2016, and tested by RT-PCR.The results showed that 24 samples were TGEV positive, and the positive rate of TGEV was 11.76%, which is higer than the national TGEV survey results in China Among the seven cities, Chifeng city with the samples of the highest infection rate, at 20.83%. The pathogen detection rate was different by reason of the types of samples, therein TGEV detection rate of anal swab was the highest with 15.56%, and the second was intestinal, and the lowest was feces, and their positive rate of TGEV was 12.00%% and 7.87%.We analyze the TGEV-CF-S gene and other 25 gene sequences from other domestic and the international strains, and found that the nucleotide homology was up to 95.1%-99.2%, and TGEV-CF (NMG) strain stay the closest relative with attenuated H and H16 from China.
