Effects And Mechanism Of ART Combining With CoSMM On Chicken Coccidiosis Infected With E.Tenella

Avian coccidiosis is a serious parasitic disease caused by E.meria that parasitis on intestinal epithelial cell. It caused great financial losses.The wide use of anticoccidial durg caused durg resistance. Therefore it is extremely urgent to research the new coccidial drug. Chinese herbal medicine was receiving much more attention on account of it’s advantages include it derives from natural, the low toxic and side-effects, less resistance.More and more have been proved possess anticoccidial effect such as dichroa,sopharate flavescentis.ART is a effective anti-malaria constituent extracted from Artemisia annua L. It is a sesquiterpene lactone contains peroxide bridge. Chinses pharmacologist Tu Youyou won the 2015 Nobal prize in medicine for her discovery of a new approach to malaria treament.ART could not resist the plasmodium, but also resist Leishmania, schistosome and coccidium. In recent years, studies have shown that ART has certain curative effect to coccidium. The co-sulfamonomethoxinum is consisted of sulfamonomethoxine and trimethoprim. They are offen used for prevention and control of bacterial infection and coccidiosis of livestock and poultry. In this experiment, we duplicate the coccidiosis model used E.tenella. Than we investigated the anticoccidial efficacy of the combination of ART and CoSMM against coccidiosis.1.The aim of this study was to study the therapeutic effects of the combination of ART and CoSMM aganist E.tenella.80 chicken were raised to 13-day-old and than divided randomly into five groups. Except the control group, the other chickens were orally inoculated with 80,000 sporulated oocysts per bird respectively. The medicines was given for chickens from one day before the inoculation to the end of the test. The efficacy of medicines was evaluated through the following items:survival rate, average daily weight gain, lesion scores, the anticoccidital indexs(ACI), the index of immunity organ after the test. Results show that the three medicines could increase the ACI and reduce lesion scores caused by coccidia infections. The combination groups showed the best anticoccidial effect and increased significantly the average daily weight gain compared with El and ART group. It suggested that the combination of ART and CoSMM had better thera peutic effect than alone.2.In order to explore the synergistic effect of ART and CoSMM on E.tenella.80 chicken were randomly divided into five groups.Except the control group, the other chicken were orally with 8×104 sporulated oocysts per bird respectively. It would be done including observation of histopathological changes in caecum by paraffin section and SEM, statistical anailsis of proinflammatory and antiinflammatory factor in the protein and gene level. The results showed that the drug treatment groups especially the combination group improve the pathological change and the content of inflammatory factors. This suggested that the combination of ART and CoSMM had synergistic effect by reducing inflammatory reaction.3.To explore the effect of the combination of ART and CoSMM on E.tenella. Apoptosis retio and mitochondrial transmembrane potential by flow cytometry. Ultra structural changes were observed by transmission electron microsocopy. The results showed that compared with the challenge control group, the amount of second-generation merozoites of other groups have decreased by 23.22%,35.49%,53.34%;the apoptosis retio have decreased by 425%、586%、 962%;red/green ratio have increased by 34.41%,25.41%,71.26%.Ultra structural of the second-generation merozoites of drug treatment group have showed apoptosis performance especially the combination group.It suggested that whether ART or CoSMM can promote second-generation merozoites apoptosis, and the combination of ART and CoSMM can faciliate this phenomenon.In conclusion,ART and CoSMM show the synergies effect on the therapeutic effect,cecal lesion,intestinal inflammation,the apoptosis of econd-generation merozoites when we use the half does of two medicine.ART indicates anticoccidial effects through alleviation of intestinal inflammation and the apoptosis of econd-generation merozoites.CoSMM indicates anticoccidial effects through the apoptosis of econd-generation merozoites.The combination of the medicians indicates anticoccidial effects through the two sides.
