| In order to better explore Sugar-beet male sterile cytoplasmic and nucleargene in regulating protein correlation. In this paper, we use the method of molecular marker technology and if it is fertile via to survey its pollen to select the S type cytoplasmic Sugar-beet with pollen,the S type cytoplasmic Sugar-beet without pollen(male sterility line)and its maintainer Iine,Then we Put forward the Leaf and Flower buds protein of them to analyze the difference btween them after Two-dimensional gel electrophoresis of protein. The results and conclusions are as follows.l.We use the method of molecular marker technology and if it is fertile via to survey its pollen to selcted 5 strains Sugar-beet with S type cytoplasmic and pollen from the experimental materials want to be selcted to be male sterility line provided by characteristic Crops research institute of Inner Mongolia academy of agricultural sciences.2.Compare two protein extraction methods,the method of TCA-acetone is btetter than Acetone-phenol in Protein extraction about leaf and bud ot Sugar-beet , Then we ameliorate the acetone TCA-method of Protei extraction.3.The technique parameters of two dimensional gel electrophoresis sugar beet protein were optimized, and the clear bidirectional protein map wa obtained.4.The 12 different protein spots has been found comparins Two-dimensional electrophoresis figure of the leaf protein between m sterility line and its maintainer line,7 of them be owned only by male sterilit line,5 of them be owned by its maintainer line.The 23 different protein sp has been found comparing Two-dimensional electrophoresis figure of the b protein between them,15 of them be owned only by male sterility line,8 them be owned by its maintainer line.5.In the paper we aslo compare the of the Two-dimension electrophoresis figure between each one of the S type cytoplasmic Sugar-bee and the S type cytoplasmic Sugar-beets with out pollen (male steri line),We respectively found 5,4,6,8,11 Differentially expressed protein spots be owned by the five S type Sugar-beets with pollen. |
