| According to Food and Agriculture Organization statistics, China’s poultry production ranking of second place in the world, after the United states. Guarantee the safety of poultry products is not beneficial to public health but also to promote the rapid and healthy development of China’s import and export trade. Antiviral drug is one of the most important risk factors for the quality and safety of poultry products. Exalted clinical using of antiviral drugs such as amantadine, ribavirin also have some preventive treatment to animal’s common viral infections, and the price is lower. Thus, it is often used as a veterinary drug to prevent and treat animal diseases, but these drugs have not been approved as a veterinary. Inappropriate use of antiviral drugs would not only lead to poisoning animals, drug residues, also can cause the emergence of resistant strains of the virus, reduce the effectiveness of antiviral drugs, which bring a potential threat to human health. Therefore, it is also necessary to develop and validate an analytical method for detecting trace residues of antiviral drugs in poultry products.This study aimed at using DSPE-LC-MS/MS to establish an analytical method for detecting 14 antiviral drugs and and relevant metabolites( including amantadine, rimantadine, memantine, moroxydine, imiquimod, arbidol, ribavirin and its main metabolite TCONH2, acyclovir, ganciclovir, famciclovir, penciclovir, oseltamivir and its metabolite oseltamivir carboxylate) in poultry products(chicken muscle, chicken liver, egg). And evaluate the purifying effect of new adsorbent(Carbon nanotubes, Graphene) and traditional sorbents(C18, PSA). The main contents and results are as follows:1. LC-MS/MS method for the 14 different polar drugs was established by optimizing the parameters of liquid phase and mass spectra. The simultaneous detection of 14 drugs was achieved by a modified C18 column Agilent SB-Aq, and 0.1% formic acid in water / methanol as the mobile phase. Moreover, the isomers rimantadine and memantine completely separated unde the optimized mobile phase gradient.2. Establish the sample pretreatment method of 14 antiviral drugs in chicken muscle, chicken liver and egg. The samples were extracted with 1% acetic acid in ACN/water(9/1), chicken muscle and liver were clean-up by PSA, egg was clean-up by C18. The recovery rates of spiked samples were 56.2%-113.4% with RSDs of 1.1%-13.9%. The LODs and LOQs for all the analytes were in the range 0.02-1.0 and 0.05-2.5 μg/kg,3. Establish the analytical method of amantadine, rimantadine, memantine, oseltamivir in poultry products using MWCNTs adsorption material. The LODs and LOQs for the analytes were in the range 0.1 and 0.5 μg/kg, and the recovery rates of spiked samples were 85.4%- 110.3% with RSDs of 1.1%-12.9%. Compared with the traditional adsorbent, MWCNTs can significantly reduce the matrix effect, and the interference of chromatographic is small. But the sensitivity of the method is not good as the traditional adsorbent.4. According to the established detection method, the samples from different regions were detected. There are some residues of amantadine, imiquimod, ganciclovir in Chicken muscle and eggs. |
