Cucumber(Cucumis sativus L.), one of the major vegetable crops in the country, is the fourth one in the area of vegetable cultivation in the world. New varieties creation using genetic engineer has become an important means to improve the yield and quality of cucumber because of the limit resources of cucumber species. In this experiment, a transformation system of glutaredoxin CsGRX1 was established and genetic transformation of CsGRX1 by somatic embryogenesis was stuied in cucumber of 14-1.1. A genetic transformation system of cucumber by the way of organ regeneration was creat, and cotyledon node was treated as explants. 0.5 mg / L Basta in MS was a half of lethal concentration. 2 d of pre-incubation, 15 min infection, 0.5 of Agrobacterium OD600 and 2 d of co-culture are optical condition for more resistant buds acquired. Adding 100 μmol / L acetosyringowie is much better in efficiency of genetic transformation.2. A regeneration system through somatic embryogenesis was established in cucumber14-1 using cotyledon node. The best callus induction medium was MS medium including1.0mg/L 2,4-D, The best differentiation medium also was MS medium including 0.05 mg/L2,4-D + 1.0 mg/L KT + 0.1 mg/L IAA.Genetic transformation of CsGRX1 was made by embryoid bodies and get the young plants.
