Through the study of glycoside aromatic substances on the inhibition Pestallozzia theae to reveal the β-glucosidase gene expression related to tea plant disease resistance, which to offer the theoretical foundation for the rapid breeding of resistant varieties.The main results are as follows:1. The tea in vitro hydroponic experiments,examined the Pestallozzia theae disease index of 19 tea varieties.The majority of tea varieties resistant to Pestallozzia theae of disease index between 9-12,the resistance of Maoxie on Pestallozzia theae of the strongest,Pestallozzia theae index minimum,6.7.And the disease index of Soubei 23.3 is much higher than other tea varieties,resistance and therefore Soubei of Pestallozzia theae should be the worst.2.19 tea varieties glycoside inhibition of Pestallozzia theae rate can be divided into 3 levels.The inhibition rate of the lowest level only Soubei a tea varieties,the inhibition rate of 73.20%,the glycosidic aroma before resistance body on Pestallozzia theae of the worst.The second level includes Xiangbolv、Wuyangxiang、Yaoshanxiulv、Xiangfeicui four tea varieties,their glycoside inhibition of Pestallozzia theae rate was 90.6-92.5.The other 14 tea varieties glycoside anti Pestallozzia theae inhibitory rate was above 95%.3. Show that the real-time qPCR experiments Wuheichangye、Xiangbolv、Zaobaijian、 Kunmingshilixiang、Mingshanzao、Tiexiang six varieties of β-glucosidase gene expression quantity is higher than Xiangfeicui,of which iron incense high Xiangfeicui up.But the other 12 tea varieties with respect to Xiangfeicui,β-glucosidase gene expression amount is low,especially the Soubei,the biggest difference.4.β-glucosides aroma of Pestallozzia theae inhibition rate experiments showed that,in addition to tea varieties,Xiangbolv inhibitory rate was 89.77%,the inhibition of the remaining 18 tea varieties are more than 90%rate.Inhibition of Maoxie tea varieties rate reached 98.35%,in the experimental group was the highest.5. GC-MS spectroscopy was used to determination of β-glucoside kind of aroma substances benzene methanol showed,tea variety Tiexiang β-glucoside kind of aroma substances benzene methanol content is much higher than other varieties of tea powder, 10g β-glucoside kind of aroma substances benzene methanol content reached 16.279μL.Soubei、 Changyebaihao two tea cultivars of β-glucoside kind of aroma substances benzene methanol respectively is 3.849μL,2.883μL,lower than the average level of.6.19 tea varieties or strains of disease index and its glucoside inhibiting body to just before Pestallozzia theae disease rate basically negative correlation,but there are still individual tea varieties or lines deviation.There was no significant linear relationship between grape and β-glycosidic aroma benzene methanol content,p-glucoside kind of Tea Aroma Pestallozzia theae inhibition rate,difference of β-glucosidase gene expression quantity,Pestallozzia theae index of tea four.Possible reasons are the following.One is the tea plant disease index,in the artificial cultivation box for tea shoots in vitro cultivation,may lead to disease index of instability.Tea plant alkaloids,such as free aromatic oil composition has inhibitory effect on Pestallozzia theae;two p-glucoside kind tea aroma,detection of β-glucosidase aromatic oil,in addition to P-glucosides aroma of benzyl alcohol,and some will have an impact on the Pestallozzia theae non grape p-glycosidic aroma components,such as amyl alcohol、fumaric acid methyl ester,The three adjacent phenol.Aromatic components in the formulated into PDA medium,would have lost due to higher temperature in different degree,the experimental results of deviation. Aromatic components in the preparation of PDA medium, adding emulsifier cosolvent, emulsifier can cause the inhibition of Pestallozzia theae.Three is the expression of β-glucosidase gene,expression of β-glucosidase gene in tea plant P-glucosidase and not with β-glucoside precursor binding, β-glucosidase aroma not only benzyl alcohol, through the literature can be seen, β-glucosides aroma and Cis-3-hexene, Phenethyl alcohol, Methyl salicylate, geraniol, linalool and four kinds of linalool oxide.
