Botrytis elliptica (Berk.) Cooke, a lily disease caused by fungi, has an adversarial impact on the growth and development of lilies. Its pathogen is Botrytis cinerea, which is able to germinate and spread quickly in the high-temperature-and-medium-humidity environment. This infection seriously impedes the centralized large-scale production of lilies, which consequently results in enormous economic losses. A variety of physiological indicators of lilies will change after they are infected by Botrytis cinerea. The Mitogen-Activated Protein Kinase (MAPK) is an essential building block of the immune system of plants, and it is closely related to the resistance to fungal diseases. We take advantage of MAPK to conduct experiments on revealing whether and how the infection of Botrytis cinerea affects lily shoots.In this experiment, we utilize the Asian lily species "Jingcui" as our experimental material. We establish the rapid propagation system by selecting the optimal culture and subculture conditions. We then determine several physiological indicators related to the correlation between the lily shoots and the biotic stress of Botrytis cinerea. In addition, a cloned MAPK gene, which is related to the resistance to Botrytis elliptica (Berk.) Cooke, is first obtained by having the testing samples infected by Botrytis cinerea, and is then analyzed using bioinformatics techniques. The results of this study are as follows:1.In the experiments of building rapid propagation system, the optimal medium of the induced scale differentiation is MS+6-BA1.0mg/L+NAAO.lmg/L. The optimum medium of scale adventitious buds subculture is MS+6-BA1.0mg/L+NAA0.1mg/L. The optimum medium for promoting aseptic root knot is 1/2MS+sucrose 60g/L+NAA0.5mg/L.2.In the experiments of analyzing the correlation between the lily shoots and the biotic stress of Botrytis cinerea, the SOD, POD, PPO, PAL activities of infected lilies (treatment group) are higher than those of normal lilies (control group). The content of soluble protein in the treatment group is lower than that in the control group, while the content of soluble sugar in the treatment group is higher than that in the control group.3.We obtained a cloned MAPK gene which is relevant to anti Botrytis elliptica (Berk.) Cooke using Jingcui lily shoots and the RACE technology. Its total length is 161 lbp, including an Open Reading Frame (ORF) with 1128bp. It encodes 375 amino acids and its molecular weight is 42.8kDa. Through multiple analysis, this MAPK gene shares similarities with Arabidopsis MAPK4 and rapeseed MAKP4.
