Actinidia eriantha Benth (A eriantha Benth) is a liana of Actinidiacea, which another name is Baishanmaotao or hairy gourd. The root of A. eriantha is usually used for civil treatment of gastric cancer, breast cancer, esophagus cancer, liver cancer et al in She nationality. However, the active compounds and mechanism is still unclear. The aim of this study is to probe the anti-cancer parts and compounds, anti-cancer mechanism of A. eriantha.In this study, we first seperated the ethanol extraction(AE-E) from the root of A. eriantha with flash chromatography to obtain the dichloromethane and methanol parts named AE-E/DM, AE-E/M respectively. Together with silica gel, sephadex column chromatography and thin layer chromatography we obtained a white power compound which identified to be 2a,3a,24-trihydroxyμrs-12-en-28-oic acid through HRES1-MS/MS and NMR technology. We measured the compoμnd content in different original material by HPLC-PAD technology, including content in each extracted parts.This research analyzed the anti-tumor potential of three crude extractions and the compound 2a,3a,24-trihydroxyurs-12-en-28-oic acid with several tumor cells through MTT, the results indicated the AE-E/DM could significantly inhibit the proliferation of liver cancer cell lines(HepG2), prostate cancer cell lines(PC3), gastric cancer cell lines(BGC823) and colorectal carcinoma cell lines(sw620). What’s more, the 2a,3a,24-trihydroxyurs-12-en-28-oic acid also showed high inhibition on the four cell lines’ proliferation. In this paper we further analyzed the in vivo anti-colorectal carcinoma activity of the compound with xenograft nude mice model. The result indicated that it could dose-dependently suppress the colorectal carcinoma growth, which was speculated to be the potential active compound of A. eriantha. This research also investigated the anti-colorectal carcinoma mechanism of this compound with AO/EB fluorescent staining, flow cytometry(FCM) and western blot. The results indicated it could induce cell apoptosis together with autophagy.This study made a foundation for the material basis which showed anti-cancer activity of Actinidia eriantha Benth and provided scientific evidence for its oncotherapy use.
