Tomatoes have the advantages of rich nutrition, wide adaptability, easy cultivation, high output, versatility and so on. So, tomato is an important vegetable in the world. It is also one of the main vegetables in our country. The plant of tomato accounted for about 40% of cultivation of vegetable area. Occupy the main position in crop of agricultural production. Especially in the north cold season tomato resistance to low temperature is particularly important for production, improve the vegetable production, promote the farmers’ income. Breeding resistant varieties of tomatoes, realize the collocation of molecular techniques and cultivation, can improve the yield of tomato in low temperature season, it is of great significance, Because of low temperature resistant tomato varieties can not only adapt to the production, adjust the off-season supplies, but also can increase the income of producers, in north China, the spring vegetables lack, In the north, the same greenhouse cultivation technique has good low temperature resistance of tomato germless resources is more important, there are some tomatoes in the cold seas on production is too low, which affect the market supply, and influence the farmers’ income. According to the market demand, therefore, breeding tomato of resistance to low temperature varieties is necessary.Tomato resistance to low temperature properties belong to quantitative character, by multiple genes system control, and genetic law is very complicated. In this study by foliar-spraying with different mass concentration ABA, taking the seedlings of tomato‘Dong Nong 11537’as test materials. Study the physiological changes and differences in gene expression research.(1) By growing the same groups of tomato seedlings of leaf spraying different concentrations of ABA, low temperature of three days after to survey three cold related physiological indexe s, analysis the significance of difference to get the optimal concentration of exogenous spraying ABA for 200 ul/L.(2) on the basis of the appropriate concentration, further discusses influence of the exogenous spraying ABA on tomato seedling resistance to cold, Results show that in 200 ul/L ABA pretreatment of tomato seedlings, with cold time prolonged, the leaf proline, soluble sugar and soluble protein content were increased, at the same time the antioxidant enzyme activity and chlorophyll content was higher than CK, and MDA content lower than CK.(3) By cDNA- AFLP to analysis of the differentially expressed genes related to cold on exogenous ABA of tomato seedlings. Divided into four groups: water + cold treatment(CK); Water(CK1); 200 ul/LABA processing(CK2); 200 ul/LABA + cold(A3), extraction of total RNA in each group under mixing time in each team, analysis on the reverse transcribed into cDNA markers, finally obtain difference banding 50, the Blast analysis that the function of these genes inclu ding signal transduction, cell metabolism, transcriptional regulation, gene synthesis and modification, etc. Some of these genes and TDF sequence plays an important role in plants.(4) in the selected part of the genetic associated with cold resistance, b y real-time fluorescent quantitative PCR verify its relationship with the ABA, by using2-△△Ct method analysis relative gene expression, TDF35, TDF37, TDF41, TDF47 expression quantity rise trend, among them, the TDF38, TDF41 after in ABA pretreatment increased expression is significantly higher than CK, it is consistent with the experiment results.
