Polygonatum sibiricum Red is a perennial herb of lily family. Mainly distributed in the north temperate zone and north subtropical zone, China has also widely distributed.Polygonatum in dried roots as medicine, the traditional breeding method is using the tuber planting, which will not only cause waste of medicinal material itself, will also increase the offspring degradation. Use the yellow seed lean breed, which can effectively solve the above problems, but also to avoid the waste of seed. Previous studies have made a lot of research on the reasons of dormancy, dormancy breaking methods, germination characteristics and propagation methods of the seeds, but the storage physiology of the seeds of. Excellent seed is the main condition of plant health, scientific storage condition is to make the seed life is prolonged, the premise that the life is strong. Different plants, the physiological and biochemical characteristics of seeds are different, in the process of storage, seed vigor and storage characteristics will show a greater difference. Different storage conditions also affect the seed vigor. In this study, we used different storage temperature, water content and different light conditions to store the seeds of Huang Jing for a period of 6 months. Of Polygonatum seed antioxidant system(SOD, CAT and POD) activity, conductivity, content of MDA content, storage substances(soluble sugar, soluble protein, crude fat) and germination rate were determined, a basic understanding of the p.sibiricum seeds during the storage period, the physiological and biochemical changes, seed deterioration process, seed vigor decreased. In order to determine the best storage conditions of the seeds of Polygonatum sibiricum Red, to provide a theoretical basis for sexual reproduction and Germplasm conservation. Research results:(1) under different temperature conditions, the seed of the yellow seeds stored in different temperature had different changes in physiological indexes with the prolonging of the storage time. 4℃ storage of seeds during the storage period of anti oxidase activity decreased the slowest activity remained the highest conductivity and MDA content was the lowest and the slow rise, after 6 months storage germination rate was 71.08%, and room temperature(temperature of 15℃-30℃) and storage of seed germination rate was 42.3%. 4℃storage of seed content of storage substances(soluble sugar, soluble protein and crude fat) changes relatively slowly, indicating that low temperature can in a certain extent, inhibited seed respiration, alleviate the seed internal metabolic activity, membrane lipid peroxidation and slow, internal storage material consumption is small, seed vigor decreased more slowly.(2) the seed with different water content of the seed with the extension of the storage time of the indicators have also changed to varying degrees. 3% water content of p.sibiricum seeds during the storage period, the activity of anti oxidase system maintained at a higher level, electric conductivity and MDA content the lowest storage material was relatively stable, occurs only very small changes, after 6 months storage germination rate was 72.5%, and 15% water content of Polygonatum sibiricum seed germination rate was only 33.2%, far less than 3% water content of seed. This is because with seed water content is low, the internal metabolic activity more slowly and the consumption of nutrients on seed is less, fewer free radicals produced and membrane system can get good protection, greatly reducing the seed deterioration process.(3) Under different illumination conditions, the changes of the physiological indexes of the seeds of the seeds stored in the storage period of the seeds were in the same condition, but the dark storage was obviously better than the natural light. The activity of antioxidant enzyme system of dark storage was higher than that of natural light. The electrical conductivity and MDA content was lower than that of natural light. After 6 months storage, the germination rate was 43.42%, and the germination rate was 34%. So, the dark condition is advantageous to the storage of the seed of Polygonatum sibiricum Red.
