Plant endophytic fungi act a variety biological function, by forming a symbiotic relationship with the host, by parting the synthesis proccess of active components and promoting adaptability to the stressed environment, and so on. The documents about endophytic fungi on seed germination, seedling growth were mostly focused on grasses species, and just few studies have reported growth promoting function of endophytic fungi from Sophora japonica, Platycladus orientalis and other woody plants.To explore the phosphate-solubilizing ability of endophytic fungi and their plant growth-promoting potential, the phosphate-solubilizing capability of endophytic fungi from Sophora japonica leaves was evaluated by the method of dissolved phosphorus cycle and molybdenum-bule, chlorophyll, soluble protein content, SOD and POD indic were detected after inoculating sterile host plantlets. The results showed that, there were 28 out of 55 fungi strains from ancient Sophora japonica leaves with phosphate-solubilizing capability, and 12 fungi among 28 displayed strong capability of dissolving phosphate of Ca3(PO4)2 by a test of phosphate-solubilizing capability, two strains with strong phosphate-solubilizing capability were then picked out up, ZG-15 and ZG-34, belonged to Aspergillus genus.Their d and d/D data were both higher than the others, and their solubilization of Ca3(PO4)2 accounted for 1238.28 mg·L-1 and 941.22 mg·L-1, respectively, which were significantly higher than the control(P<0.01). Four strains(ZG-7, ZG-9, ZG-15 and ZG-48) from different genus were inoculated into aseptic Sophora japonica seedlings bottle to ensure their contact upon with each other. Mycelium was observed infecting into the cortex cells and around the surface epidemical cells of root tissue; under the treatment by Aspergillus niger ZG-15, Sophora japonica seedlings’ Chl. content, soluble protein content, SOD and POD net activity index were all obviously higher than the control(P<0.05), ZG-15 can maintain healthy seedling growth and improve seedling resistance, which showed a potential for conservation of ancient Sophora japonica and dispalyed a potential application as an organism-fertilizer for forest tree.To explore functional endophytic fungi for salt tolerance of Sophora japonica, eight strains were inoculated into the Sophora japonica seedlings with 200 mmol·L-1 NaCl stress by root-irrigation. The seedling height, root-shoot ratio, dry weight, water content, chlorophyll content, Pro content, MDA content, SOD and POD activities were detected, respectively. The results showed that, under salt stress, the seedlings’ biomass and physiology changes were basically same. Isolats H-1、H-3、H-4 and H-6 could improve the salt resistance of Sophora japonica seedlings, H-5 were just the opposited, and the others were not obvious. The results showed H-1, H-3, H-4 and H-6 were the dominant functional isolates among the tested endophytes, and they all showed potential application in practise.To explore functional endophytic fungi for promoting seed germination and seedling growth of ancient Chinese arborvitae, eight strains obtained from ancient Platycladus orientalis leaves were re-inoculated into the seeds and seedlings of arborvitae by seed soaking and root-irrigation, the seed germination ratio, germination potential, germination index, and seedling height, ground diameter, root- shoot ratio, dry weight, water content and chlorophyll were detected, respectively. The results showed that, Epicoccum nigrum C-4 significantly promoted the arborvitae seed germination and seedling growth. The germination potential, germination index, and seedling height, ground diameter, dry weight, water content and chlorophyll a/b were all significantly higher than that of the control by C-4 suspension. Aspergillus fumigatus C-6 just promoted the growth of the seedlings. Alternaria C-5 showed obvious inhibiting effect on seed germination and seedling height. Trichoderma harzianum C-1 only showed obvious inhibiting effect on seedling height by root-irrigation of C-1 suspension.Whereas, the other tested strains displayed unsharpness roles in this experiment. The 8 strains acted promotion, inhibition, or resultless roles on arborvitae seed germination and seedling growth, C-4 and C-6 were the dominant functional strains among the tested endophytes with potential function for ancient Platycladus orientalis protection and regeneration.
