| [Objective]Study proanthocyanidins whether to protect zearalenone causes damage in mouse liver and kidney.[BackgroundJZearalenone have special estrogen toxicity.and widely present in cereals.and recent studies on the reproductive toxic ity of zearalenone relatively large,and study its oxidative damage toxicity less;Procyanidins have antioxidant effects.and a wide variety of sources,Therefore, according to this trial of zearalenone oxide toxicity.Select antioxidant proanthocyanidins,To study whether the protective effect of procyanidins against of zearalenone cause oxidative damage.[method]Select a clean healthy male Kunming mice 40,randomly divided into four groups,They are:The control group (saline i.p.),Procyanidins group (100mg/kg PC i.p.).Zearalenone group (40 mg/kg ZEA i.p.),Proanthocyanidins+Zearalenone group (100 mg/kg PC i.p.,40 mg/kg ZEA i.p.),Continuous administration of 7 d,After the last dose in fasted water,8h after were sacrificed.Collection of blood and liver.Measurement of blood and biochemical indices of the liver.Detecting blood UA, BUN, AST, ALT.Liver tissue MDA, GSH-Px content and T-SOD activity.Detecting liver nuclear factor E2-related factor 2 (Nrf2), glutathione peroxidase (GSH-Px), heme oxygenase 1 (HO-1), y-glutamyl cysteine synthetase (γ-GCS), quinone oxidoreductase 1 (NQO1) gene and protein expression levels.[Results] Compared with the control group,Zearalenone group of mice blood UA, BUN content was significantly increased (P<0.01,P<0.01),ALT and AST were significantly increased in the blood (P<0.01, P<0.01).Liver T-SOD, GSH-Px activity decreased (P<0.05, p<0.05),MDA content significantly increased (P<0.01);Proanthocyanidins group T-SOD and GSH-Px activity was significantly increased (P<0.05),MDA content compared with control group was not significant (P> 0.05).Proanthocyanidins+Zearalenone group compared to the Zearalenone group,Mouse liver tissue content of GSH-Px, T-SOD activity significantly increased (P<0.05, P<0.05),MDA content was significantly reduced (P<0.05).Compared with the control group, Nrf2, GSH-Px, HOI, γ-GCS, NQO1 gene and protein expression levels of Zearalenone were significantly reduced (P<0.01, P<0.05).Compared with Zearalenone group,Proanthocyanidins+Zearalenone group, Nrf2, GSH PX, HO-1, y-GCS, NQO1 gene and protein expression levels increased significantly (P<0.01, P<0.05).[Conclusion] Zearalenone can cause oxidative damage to the liver of mice, reducing the expression of Nrf2 and its downstream mRNA and protein;Procyanidins can enhance the antioxidant capacity of the liver of mice, and increase the expression of Nrf2 and its downstream mRNA and protein;Procyanidins reduce oxidative damage caused by zearalenone on mice liver, play a protective role. |
