Taurine is one of the most abundant free amino acids in the mammalian body, its physiological function is very wide, and it plays an important role in the function of the organ and tissue. Taurine not only can be synthesized in the body by itself, but also obtain through food.The key enzymes of taurine synthesis are cysteine dioxygenase (CDO) and cysteine suLfinate decarboxylase (CSAD/CSD), the synthesis of taurine by CDO-CSD pathway is an important way for the synthesis of taurine in vivo.Taurine can be transported to intra-celluLar through taurine transporter (TAUT) to maintain its concentration.At present, there is still lack of relevant information about the synthesis of taurine in pigs and its effect on pigs. In this study, the expression of CDO and CSD and TAUT and the content of taurine in Landrace liver, hypothalamus, pituitary, thyroid, adrenal and testis were detected by the way of RT-PCR, Western-blot and ELISA. The resuLts will lay a foundation for further study on the effect of taurine and its application in ovis aries production.The resuLts proved that:1. CDO cDNA coding sequence in Landrace liver tissue is 609 bp, encoding area is in 1-603 bp, and encods 201 amino acids. The nucleotide sequence of CDO cDNA in Landrace liver has 99% homology with that of boar liver which was already known, and the putative encoded-amino acid sequence was exactly the same as that in boar liver (100%).2. The expression of CDO, CSD and TAUT mRNA and protein were detected in 7 tissues of Landrace. The CDO mRNA expression levels in tissues from high to low are:testes, ovaries, thyroid, liver, pituitary, hypothalamus and adrenal; CSD mRNA expression levels in tissues from high to low are:thyroid, pituitary, ovary, liver, hypothalamus, adrenal, testis; TAUT mRNA expression levels in tissues from high to low are:pituitary, ovary, liver, hypothalamus, thyroid, adrenal, testis.3. The content of taurine in serum and tissues from high to low order are:pituitary, hypothalamus, adrenal gland, liver, testis, ovary, thyroid gland and serum.The resuLts indicated that:Taurine is a widespread amino acid in pigs. Landrace hypothalamus, pituitary, adrenal gland, liver, testis, ovary and thyroid can bio-synthesize taurine by CDO-CSD pathway, but the concentration of biosynthesized taurine can not meet the demand of each organ function, it also need obtain from the blood by TAUT transportation. Taurine may plays important role in hypothalamus, pituitary, adrenal gland, liver, testis, ovary and thyroid function of pigs, but the specific function need to be further investigated.
