Liaohe estuary wetland located in Panjin City, Liaoning Province, as a national nature reserve,about 30 kilometers away from the city, located in the Bohai Bay, Liaoning top Liaohe river into the sea, with a total area of 128000 hectares, is the best preserved in the world at present, the largest and most complete ecological vegetation plots.Liaohe estuary wetland (July 20,2015,approved by the State Council, Liaoning Liaohe Estuary National Nature Reserve officially changed its name to the Liaoning Liaohe River Estuary National Nature reserve. Therefore, wetland changed its name) reed varieties are different, widely distributed, has been generally accepted in local self-generated and imported a total of about a dozen varieties. However, due to the environmental impact and lax management, reed resources are increasingly messy. Study on the classification of reed not only for exploring the reed origin, phylogeny and evolution rules, but also identifing the complicated reed plants to species or varieties, and according to the system arrangement, it is convenient for people to understand and use of plant.To clear the area of reed variant types, of Intervarietal variation, this study employ Liaoning Province reed Institute senior expert guidance, the choice can be clearly identified and publicly reported 11 reed variants as the object of study, from two aspects of the morphology and genetics into morphological difference analysis and genetic variation analysis, in order to reveal phenotype diversity and genetic diversity of reed variations in this area.To research the absorption of different varieties of reed magnesium ion in the wetland system,using the pilot unit for papermaking wastewater irrigation.Analysis under duress of heavy metals magnesium, each reed variants response and performance,and it is of great significance for focusing on cultivating strong resistance, with excellent traits reed new varieties, improving reed production and achieving reed industrial production and the absorpting of water quality, protecting the habitat.To investigate the morphological variability of eleven reeds varieties, this study focused on field investigation and statistical methods to compare morphological indices including basal diameter, height of aerial stem, leaf length at middle stem, leaf width at middle stem, internode length at middle stem, number of internodes, and panicle length in Liaoning Province Liaohe Estuary wetlands. The results show that:(1)Seven morphological indicators of reed phenotype are significantly different in Liaohe Estuary wetlands. The order of this 7 morphological indicators of intraspecific variation is panicle length> internode length at middle stem> leaf length at middle stem> leaf width at middle stem> basal diameter> number of internodes> height of aerial stem; (2)The order of the morphological variation between reed varieties is Light brown reed>Pink ear reed> White ear small reed>Coloured reed>Zhaoquanhe reed>Tight ear reed>Vertical purple reed>White ear reed> Phragmites hirsuta>Panjing reed>Small purple loose ear; (3)The principal components analysis of the seven indexes of reed varieties shows that the cumulative contribution rate of the first three principal components is to 86.001ï¼…, in these three principal components,the height of aerial stem,the basal diameter,the panicle length and the number of internodes has the biggest difference in the morphology between reed varieties,which are the main factors that lead to the phenotypic differences among the reed varieties in Liaohe estuary wetland; (4)Based on clustering analysis, Liaohe Estuary wetlands reeds can be divided into five categories, which are Phramites hirsuta, pink ear reed, vertical purple reed, white ear small reed, and a fifth large class composed of coloured reed, tight ear reed, Zhaoquanhe reed, light brown reed, small purple loose ear, Panjin reed and white ear reed. This analysis shows that there are significant differences in morphology among Kou reeds and all the differences result in a wealth of phenotypic diversity in Liaohe Estuary.This test analysis 11 reed varieties of plant total DNA genetic variation using ISSR molecular markers, according to Columbia University Canada released by ISSR primers sequences, synthesized by Shanghai biological engineering company, according to determine the reaction system, based on the article from 100 ISSR primers amplification, filter some primers that can be obtained clear bands, stable response of 18 primers for further analysis. ISSR - PCR reaction results show that:(1)In the genetic variation analysis of the 11 reed varieties using ISSR molecular marker technology, we screen out 18 primers that has good polymorphism and distinct bands from the 100 ISSR primers and the DNA sample of the 11 reed varieties obtain 136 amplification sites including 115 polymorphic sites. Each primer has an average of 7.6 amplification sites with the percentage of polymorphism reaching up to 84.2ï¼…, (2)Though clustering analysis with UPGMA, we find out that the 11 reed varieties can be divided into 8 groups in which the first group has 4 varieties, including panjin reed, small purple loose ear, tight ear reed and vertical purple reed, and the rest of the 7 groups has only one varieties in each group respectively, white ear reed, zhaoquanhe reed, coloured reed, pink ear reed,white ear small reed,phragmites hirsuta and light brown reed. Among the 11 reed varieties, panjin reed and small purple loose ear have the closest genetic relationship and light brown reed has the most distant geneti relationship with the rest of the 10 varieties. So there has been a rich genetic diversity among the reed varieties in Liaohe estuary wetlands.From the Liaohe estuary wetland reed variant absorption function of magnesium ions obtains,in 11 reeds variants, the strongest of absorption magnesium ions is pink ear reed,the weakest is Phramites hirsuta. Papermaking wastewater irrigation helps reed absorbing magnesium ions in wetland system,and the higher of waste water concentration, the stronger its absorption.
