| Cordyceps militaris is one of the most popular nutraceuticals in Eastern Asia. It has been used as a substitute for Ophiocordyceps sinensis in traditional Chinese medicine and health foods because of their similar chemical composition and medicinal properties.Cordyceps militaris contains a broad range of bioactive compounds. Cordycepin, i.e. 3 ’deoxyadenosine, is the first isolated nucleoside antibiotics from the fungus,which play animportant variety of pharmacological effects in anti-tumor, anti-bacterial, anti-viral, immune regulation, scavenging free radicals, etc.Cordycepin has been tested in the treatment with leukemia in the clinic and has a good prospect for clinical application. At present,the main source of Cordycepinis Cordyceps militarisand cordycepin was listed as the enterprise standard by many companies.Although artificial cultivation of C. militarisfruiting body has been industrialized, but the product quality andeffect is diversed. Under Chinesehealth strategy, more and more companies concerned about the health effects and cordycepin in products, and the current test data showedthat the cordycepin content in the vast majority of C. militarisfruiting body is extremely low.In this paper, we improved the dection of the cordycepin in processing time and optimized the medium for fruitbody cultivation. We also discussed the influences of different additives for cordycepin in the medium. Comparison of major bioactive compoundsof C.militaris fruiting bodies cultured on wheat medium and pupae. We detected ordycepin content in different stage of fruiting bodies and the expression level of cordycepin biosynthetic gene. The results are as follows:1. The sample treatment method of cordycepin of standard method for the determination by the Ministry of Agriculture was optimized.The results showed that the optimizedextractiontime is 30 minutes,significantly better than the original standard extraction time of 3 hours, andenergy and time-saving.2.Optimizing medium by mixing different concentration of pupa powder and found the best concentration is 10%. In this concentration, we found that the best result is wheat mixed the eggsfollowed by wheat, and maize is lowest. Considering the effect and cost of mild powder,as well as the influence of cordycepin content mixed the additive, we can get the best result when the concentration is 3g/L.Peptone concentration changed from the original 10g/L to 6g/L.They have the same effect,andthere was no significant difference in the biostatistics.3.Adenosine and triacontanol has played a positive role related on increasing the content of cordycepin, the optimum concentrationis2mg/m L and 3mg/Lrespectively.Iron and manganese ions can promote cordycepin production, the optimum concentrationis0.05% and 0.01%respectively.With the optimum concentration of manganese ions adding, cordycepin content can reach 1.4 times than the control group.4.C. militaris that was cultured on pupae was divided into three types of samples, sclerotium, pupae FB and a mixture of the two. The content of bioactive compounds, were compared among the wheat FB and the aforementioned three types of samples. The pupae FB have the highest cordycepin content, the most important bioactive compound in C. militaris. The sample of sclerotium has the lowest content of all the bioactive compounds, except for the nitrogen content. Comparing the fruiting bodies cultured on pupae and wheat, pupae FB has higher contents of cordycepin, carotenoid, SOD and nitrogen than those of wheat FB. Considering the adenosine, polysaccharide and mannitol contents, the wheat FB has higher levels. The mix and wheat FB did not have significantly different cordycepin, carotenoid and SOD contents. The nitrogen content in the mix was higher than that of the wheat FB. The adenosine, polysaccharide and mannitol contents in the mix were significantly lower than those of the wheat FB. |
