Learning and memory is one of the major central nervous activity, and the mechanism is very complex and diversified. Learning and memory mechanisms occur by variety of signaling proteins,and eventually form a complex signal transduction network. For signal transduction proteins and knowledge of the signaling pathway, which will facilitate learning and memory-related diseases,and basic molecular mechanisms of drug targets theory of learning and memory impairment, and therefore, from the theoretical and practical value in terms of a very important significance. So far,narcotic drugs and perioperative factors on ancillary drugs on learning and memory in signal transduction mechanisms which was not reported in systematic theoretical research. Clinical commonly used drugs such as ketamine, as there is no research, thus research in this area is very necessary and its role and mechanism of the correct application of the drug clearly contribute to the future provide the basis.We select 39 young male Wistar rats, weighing 220-240 g, which were randomly divided into13 groups for the group C, K20 group, K80 group, CY group, hyperthermia group; intraperitoneal injection of ketamine methods in administration 10 min, 6h, 10 h after by Western Blot to detect the hippocampus ARC, CREB, p-CREB, ERK p-ERK protein expression and to detect changes in the hippocampus ARC, CREB, ERK mRNA by RT-PCR, to speculate drugs on ERK signaling pathway impaction.Through testing, we found that after injection 10 min, both the low-dose group, addicts high temperature or anesthetic dose group, their ARC, protein expression of p-CREB compared with the reference group were all increased in and the same trend, however, the expression of p-ERK proteins do not conform to this trend. After administration 6h, our protein expression in the hippocampus of the analysis, the amount of stimulant group, addicts and narcotic dose group compared with the control group, the protein index were increased, and had the same trend. 24 h,change the amount of each protein expression after administration of exactly the same trend, and the low dose group at 24 h after dosing, significantly higher expression of ARC, while the protein expression of addiction and narcotic dose group was significantly lower than the control group. At the same time, by Real Time PCR results show, ARC, CREB, ERK mRNA in the trends of each period, and Western Blot protein expression of the basic line, which confirms the reliability of the experimental results.The results show that the high temperature caused by doping amount of ketamine caused a transient increase in memory, anesthetic doses of ketamine can cause short memories rise, after a long memory decline results, long-term injection of stimulants can lead to decrease the amount ofketamine memory levels. |
