On the base of phenological,107 dateplum persimmon(Diospyros lotus L.) genetic resources were observed morphologically; according to the method of linkage between groups, cluster was carried by DPS and NTSYS software. Ralationship and genetic diversity of 47 female plants and60 staminiferous persimmon genetic resources have been researched by SSR molecule marker technology combined with clustering analysis; Research on germplasm resources and molecular marker; therefore their genetic relationship and differences were analyzed. Tis study achievev the following main results:(1)Observation the fruit longitudinal diameter,fruit fruit equatorial diameter,Fruit shape index,assignment,seed weight,fruit color,leaf color and flower bud,antherand pollen on 47 female plants and 60 staminiferous. Different dgrees of diversity exhibited in differentstraits.Variablecoefficient reached 11.05%,12.35%,11.96%31.25%16.24%,31.25%,10.78%,14.84%,11.71%, and 10.17%.There is a big difference between the morphological description of dateplum germplasm. In addition, each bud of female plant, pollen, anther aspect ratio of the value of fruits and seeds and male plants of cluster analysis, the results of further explanation, there is a big difference in traits.(2)The optimal SSR-PCR reaction system was establish for dateplum. In a 20μL reaction system contained 10×PCR buffer 2.0μL,template DNA 50 ng,Mg2+ 2.5mmol/L 1.5μL,d NTPs0.25mmol/L 1.5μL,Taq DNA polymerase dosage 5U/μL 0.4μL,primer 1μL,with residual volume supplied by double distilled water.(3)Female plant screened out 25 primer combinations and staminiferous plant screened out20 p rimers combinations of distinct and rich in polymorphism from 50 primer combinations.Each strain within the scope of the genetic similarity coefficient was ranged from 0.4850 to 0.8149 for strain A,0.5808 to 0.8149 for strain B,0.1785 to 0.5013 for strain DΙ,0.6154 to 0.8269 for strain DIII,0.5865 to 0.8077 for strain E,0.6058 to 0.7692 for strain F.The results show that all kinds of groups have different characters.(4)The phylogenetic tree of 47 female plants and 60 staminiferous were constructed by UPGMA method. The result show that the genetic similarity coefficient and genetic distancebetween various ethnic groups, analyzed the relationship between genetic distance between each plant, obtained dendrogram showed that clustering results between the two groups showed no significant, Diospyros between sub-plant there is a wealth of genetic differences. Female plants at the level of similarity coefficient of 0.67, can be Diospyros lotus 47 female plants into plant groups A, B, C categories, male plants at 0.662 similarity coefficient level can be Diospyros lotus60 single male plants strain into groups D, E, F categories.
