This article aimed to study the effect of swainsonine from Oxytropis glabra DC. on toxic actions and brain cell in sheep. The research contents were divided into the following 3experiments.Experiment 1: The Toxicity Attack Test of Oxytropis glabra DC. in Karakul SheepThis study was conducted to verify the damage effect of Oxytropis glabra DC.. on tissues and organs in Karakul sheep, with an aim to provide reference of studying the antioxidant indicesprobiotics. Twelve Karakul sheep with an average body weight of 16.00 ± 1.40 kg were divided into 2 groups, with 6 in each group and each sheep as one replicate, and fed one of the following 2 diets: basal diets(Control group) or basal diets supplemented with 400 g dry powder of Oxytropis glabra DC.(Test group) per day per sheep. The experiment lasted for 42 days, the body weight and blood samples were collected every 7 days,the body weight and blood samples were collected every 7 days, and the sheeps were slaughtered to provide the tissue at 42 d, and determined the serum index and tissue pathology using the automatic biochemical analyzer and hematoxylin-eosin staining, respectively. The results showed that the typical clinical symptoms and pathological change of tissues and organs were observed in Test group, the contents of AST(P<0.01), ALT(P<0.01), AKP(P<0.01), BUN(P<0.01),GLU(P<0.01) and TC(P<0.01) were increased and the content of TG was decreased(P<0.01)compered with the control group. In conclusion, the Oxytropis glabra DC. could cause lesion and damage of tissues and organs in Karakul sheep.Experiment 2: Effect of Oxytropis glabra DC. on Antioxidant Indices of Plasma and Brain Tissue in Karakul SheepThis study was conducted to investigate the effect of Oxytropis glabra DC. on antioxidant indices of plasma and brain tissue in Karakul sheep, with an aim to provide reference of improving the mechanism of toxication of Oxytropis glabra DC. Twelve Karakul sheep withan average body weight of 16.00±1.40 kg were divided into 2 groups, with 6 in each group and each sheep as one replicate, and fed one of the following 2 diets: basal diets(Control group) or basal diets supplemented with 400 g dry powder of Oxytropis glabra DC.(Test group) per day per sheep. The experiment lasted for 42 days, the body weight and blood samples were collected every 7 days, and the sheeps were slaughtered to provide the brain tissue at 42 d. The antioxidant indices of plasma and brain tissue were determined using a UV-VIS spectrophotometer and ELIASA, respectively. The results showed that compered with the Control group, the activity of SOD in plasma was decreased significantly at 15 d(P<0.05) and was decreased highly significantly from 21 d to 42 d(P<0.01) in Test group.The content of GSH in plasma was decreased significantly at 8 d(P<0.05) and was decreased highly significantly from 15 d to 42 d(P<0.01) in Test group. The content of MDA in plasma was increased highly significantly from 15 d to 42 d(P<0.01) in Test group. The contents of TSH and NPSH in plasma were decreased significantly at 21 d(P<0.05) and was decreased highly significantly from 28 d to 42 d(P<0.01) in Test group. The contents of AMA and NPSH in plasma were decreased significantly at 21 d(P<0.05) and was decreased highly significantly from 36 d to 42 d(P<0.01) in Test group. The contents of NO in plasma was increased highly significantly from 21 d to 42 d(P<0.01) in Test group. The contents of ET-1in plasma was increased from 15 d to 21 d(P<0.05) and found no significant difference at 36d(P>0.05)in Test group. The content of CGRP in plasma was increased significantly at 21 d(P<0.05) and was increased highly significantly from 28 d to 42 d(P<0.01) in Test group. In brain tissue, the activity of SOD and the content of GSH were lower in Test group than that in Control group(P<0.01), and the content of MDA, NO and ET-1 were higher in Test group than that in Control group(P<0.01). In conclusion, the Oxytropis glabra DC. could cause the decrease of antioxidant capacity and damage of tissues and organs in Karakul sheep.Experiment 3: Effect of Swainsonine from Oxytropis glabra DC. on AQP1 and AQP4 of Astrocyte in Rat.This study was conducted to investigate the effect of Oxytropis glabra DC. on toxicactions of astrocyte and aquaporins in Rat, with an aim to provide reference of relationship between swainsonine from oxytropis and encephaledema. The astrocyte were culrured by cell-culture medium containing different concentration of swainsonine(0, 0.5, 1.0, 2.0 mg/m L) for 3h, 6 h, 12 h and 24 h, respectively. The activity and survival rate of cell and the relative expression of AQP1 and AQP4 were determined using MTT and Western Blot,respectively. The results showed the survival rate of cell were decreased with increased dose and action time. Compered with the Control group, the groups added swainsonine all increased the relative expression of AQP1 and AQP4(P<0.05), while the relative expression of AQP1 and AQP4 were increased highly significantly in 1.0 mg/m L SW group and 2.0mg/m L SW group(P<0.01). In conclusion, the swainsonine from Oxytropis glabra DC. coule decrease the survival rate of cell and regulate the expression of aquaporins.
