The colour of tuber’s skin from one of cultivars Mira plant growing in greenhouse is both yellow (normal) and red. The project team sorted propagation separately on the condition of red potato skin as a filter and obtained a series of variation of red skin potato clones successfully. At the same time by stripping stem tip, removal of material culture poison plantlet. In this study, we described the source of somaclonal variation in strains in detail and did SSR analysis on them with a female Mira to identify its late blight resistance, doing the nutritional analysis. The results are as follows:(1) Red potato skins as index after several generations of asexual reproduction, obtainer RSY17 somaclonal variation of new material screening, by stripping the shoot tip culture obtained RSY17 plantlets in vitro.(2) By doing SSR analysis on Mira and RSY17 plantlets, the results indicated that among the synthesized 124 pairs of primers,35 pairs of primers did not get RSY17’s and Mira’s different polymorphic bands,89 pairs of primers have get RSY17’s and Mira’s different polymorphic bands.124 pairs of primers for 963 bands, an average of 7.76 bands per primer, Mira and RSY17’s Genetic Similarity (GS) was 0.77, Genetic Distance (GD) was 0.23. The results reveal RSY17 and their female Mira have the kinship of both commonality and difference on the molecular level.(3) Using the method of comparative physiology, we choose Favorita, BashulO and Mila which are three different varieties of potato late blight resistance as test materials, and identify the late blight resistance of new material RSY17 by measuring the necrotic area of leaves and chips which are inoculated with late blight pathogen at a certain time and the variation of antioxidant enzyme activity of the cells before and after vaccination. The results showed that the leaves’ necrotic area of RSY17, Mira, BashulO and Favorita was 2.77%,8.33%,59.20% and 85.67% respectively after inoculation 48h; after inoculation 60h, the chips’ area of RSY17, Mira, Bashu 10 and Favorita was 0.23%, 8.50%,49.80% and 91.67% respectively. As time increases, the necrotic area of leaves and chips of RSY17 developed slowly and showed the trend of stopping gradually. By comparison, the necrotic area of leaves and chips of the susceptible Favorita developing rapidly after inoculation almost reached the entire area. It follows RSY17’s late blight resistance is very high.(4) At first, we measured the contents of vitamin C, dry matter, starch, sugar, crude protein and crude fiber six kinds of nutritional quality in fresh potato of somaclonal variation new materials RSY17 and itsfemale Mira, then stored them under conventional cold and measured the content of Vc, dry matter, starch and sugar respectively in this period. The results showed that RSY17 have significant increasein the contents of dry matter, starch, sugar and crude fiber, while there was no significant difference in the content of Vc and crude protein; the tubers’ content of RSY17 and Mira after storage showed highly significant difference in Vc and significant difference in sugar, while showed no significant difference in dry matter and starch. From the absolute value of each of nutrients of fresh potato tubers and after storage, most of the nutrition indicators of RSY17 are higher than Mira, only sugar is below Mira. It shows RSY17 have comprehensive advantage in fresh and processed potato flour, starch aspects than Mira. |
