Two hundred and forty one-day-old Avain broilers were divided into four groups, and fed with control group diet (containing 0.332mg/kg Se determined by Atomic absorption spectrophotometry), AFB1 group diet (0.3mg/kg AFB1),+Se group diet (0.4mg/kg Se as sodium selenite were supplemented in the control group diet) and AFB1+Se group diet (0.4mg/kg Se as sodium selenite were supplemented in the AFB1 group diet) for 21 days. For investigating the effects of Se counteracting the toxicity of AFB1 on kidney, histopa-thology, ultrastructural changes, biochemistry, flow cytometry and qRT-PCR were used. Results were as follow:The relative weight of kidney in AFB1 group was increased, when compared with control group. Histopathologically, the major lesions of kidney included cell swelling and necrosis of renal cells in AFB1 group. By electron microscopy observation, the increased lipid droplets and expanded endoplasmic reticulum were appeared in epithelial cells in AFB1 group. Compared with control group, the content of serum creatinine and uric acid in AFB1 group were increased, while the activity of renal Na-K ATPase was decreased through biochemical method. The percentage of apoptotic renal cells was increased by FCM, the contents of Caspase-3 and Bax expression were increased, the content of Bcl-2 expression was decreased by qRT-PCR in AFB1 group, when compared with control group. Through cell cycle detection, the percentage of renal cells in G0/G1 phase was increased, and the PI value was decreased. By immunohistochemical method, the PCNA expression was significantly decreased. Compared with control group, there were no significant change of the relative weight, pathological change of kidney and changes in renal function in+Se group. Ultrastructural changes were structural abnormalities of mitochondrias and slight increase of apoptotic renal cells. The renal cells were arrested in G0/G1 phase and cell proliferation was decreased. However, compared with AFB1 group, the relative weight of kidney was increases, pathological and ultrastructural lesions, and renal functions were alleviated, the incidence of apoptotic renal cells, the contents of Bax and Caspase-3 ex-pression was reduced, the contents of Bcl-2 expression was increased, the cell cycle blockage of renal cells in G0/G1 phase and the suppression of cell proliferation were im- proved in AFB1+Se group.Conclusion:Sodium selenite supplied into the diet containing 0.3mg/kg AFB1 could alleviate pathological and ultrastructural lesions in kidney, protect renal function, de-crease the percentages of apoptotic cells, and increase the cell proliferation through ameli-orating renal cell arrested in Go/G1 phase and increasing the expression of PCNA. Sodium selenite could relieve the impairments of kidney caused by AFB1, which maybe due to its regulation on cell cycle arrest and apoptosis.
